Embodiment 1

[0053] Example 1: Inhibition of preadipocyte differentiation

[0054] Adipose-derived mesenchymal stem cells (ADMSCs), i.e., preadipocytes, were cultured and differentiated into adipocytes according to established protocols. Briefly, adipose-derived mesenchymal stem cells were obtained from ATCC (PCS500011 TM , Manassas, VA), and at 37°C in 5% CO 2 medium growth kit ( PCS500040 TM ) in the mesenchymal stem cell basal medium ( PCS500030 TM ) cultured. To induce differentiation, when the ADMSC culture grew to 70%-80% confluence, it was replaced with adipogenic differentiation medium ( Adipogenic Differentiation Kit; Gibco Life Technologies TM ,U.S). Medium was changed twice a week.

[0055] After 9 days and 15 days, the cells in the plate were partially and completely differentiated into adipocytes, respectively. Both partially and fully differentiated adipocytes accumulated lipid droplets. On day 15 post inoculation, adipocytes were treated with vehicle or with la...

Embodiment 2

[0059] Example 2: Body Fat Reduction

[0060] The ability of liposome-encapsulated latanoprost to reduce fat deposition in vivo was tested by direct injection into the inguinal fat pad of mice.

[0061] Inject 100 μL vehicle, 100 μL latanoprost solution containing 100 μg latanoprost, 100 μL EggPC-latanoprost containing 100 μg latanoprost in one inguinal fat pad of each of three animals Or 100 μL of POPC-latanoprost containing 100 μg latanoprost. Seven days later, the mice were sacrificed and the weights of the injected inguinal fat pad and the contralateral non-injected fat pad were measured. see results image 3 .

[0062] A single injection of free latanoprost did not result in a decrease in the weight of the inguinal fat pad 7 days after injection compared with the contralateral uninjected inguinal fat pad. In fact, fat pad weight increased after latanoprost injection.

[0063] Surprisingly, 7 days after injection of EgPC-latanoprost, the weight of the injected inguina...

Embodiment 3

[0064] Example 3: Short-Term Reduction of Adipocyte Counts in the Intracone Region

[0065] Free latanoprost, EggPC-latanoprost and POPC-latanoprost were respectively introduced into the orbit of rats by retrobulbar injection to determine their effects on intrapyretic fat deposition. Injection of deoxycholic acid (DCA; ) as a positive control. DCA has been approved by the U.S. Food and Drug Administration for the reduction of fat cells in the jaw.

[0066] Each of six rats was treated with monocular injections. The contralateral eye of each animal was not injected and served as a negative control. The doses and injection volumes of the animals in each group are shown in Table 1 below.

[0067] Table 1 Experimental group

[0068]

[0069] On day 7 after injection, the number of adipocytes present in the intraconical fat deposit was counted by computer analysis of histopathological images and compared with the number of adipocytes in the contralateral non-injected eye. ...

Abstract

A method for reducing a subcutaneous fat deposit or a visceral fat deposit in vivo by contacting the fat deposit with a composition of latanoprost encapsulated in a liposome formed solely of egg phosphatidylcholine (EggPC) or palmitoyloleoyl phosphatidylcholine (POPC). Also disclosed are methods for treating steatoblepharon, proptosis, and obstructive sleep apnea associated with excess upper airway fat by injecting into the eyelid, intraconal space, and upper airway fat deposit, respectively, the composition of latanoprost encapsulated in an EggPC or POPC liposome.

Description

Background technique [0001] There are several medical conditions that require reducing the amount of fat in an anatomical site. [0002] Steatoblepharon is the drooping of the front part of the fat pad of the eyelid, commonly known as eyelid bags, resulting in puffy eyes. The disease is treated with surgery, known as blepharoplasty, to remove fatty deposits from the eyelids. Surgery may have side effects such as infection, bleeding, dry eyes, difficulty closing eyes, visible scarring, eye muscle damage, skin discoloration, need for follow-up surgery, temporary blurred vision, and in rare cases, blindness. [0003] Exophthalmos, which is a forward movement of the eye relative to its normal position in the eye socket, is a more serious condition. The most common cause of exophthalmos is thyroid eye disease, an abnormal function of the thyroid gland that causes the muscles, fat, and other tissues around the eyes to swell. If left unchecked, exophthalmos can cause corneal damag...
