[0099] 94826 C140-variant group 1 / genotype 5 avian reovirus

[0100] Attenuated avian reovirus 94826 C140 was obtained by passage 140 times in chicken embryos. Attenuated avian reovirus 94826 C140 was deposited under the patent deposit name PTA-126077 on August 13, 2019 at Attenuated avian reovirus 94826 C140 is also on file as PDRC reference number: 121116 in the virus bank of the Poultry Diagnostic and Research Center (PDRC) at the University of Georgia, 953 College Station Road, Athens, Georgia 20602. Example 3 describes in more detail the safety and efficacy of 94826 C140 as a live attenuated reovirus vaccine.

[0101] The parental 94826 avian reovirus is a group 1 / genotype 5 avian reovirus and is described in more detail in International Application No. PCT / US2015 / 013449 (WO 2015 / 116778) and US Patent Application No. 15 / 223,623 , which are incorporated herein by reference.

[0107] Live attenuated reovirus vaccine candidates 94826 C140 and 96139 C140 isolates

[0108] 94826 C140 avian reovirus is a variant group 1 / genotype 5 avian reovirus, as described in Example 1 .

[0109] 96139 C140 avian reovirus is a variant group 2 / genotype 1 avian reovirus, as described in Example 2. 96139 C140

[0110] embryo passage

[0111] Both virus isolates were serially passaged in specific pathogen-free (SPF) embryos via the allantochorionic (CAM) inoculation route. Specifically, CAM from the original isolate was diluted 1:100 in sterile minimal essential medium (MEM) and 0.1 ml was inoculated into CAM of 9-11 day old SPF embryos. At 48 hours post-inoculation, embryos were refrigerated at 4°C for 4 hours, and CAMs were collected from embryos, mixed, homogenized in viral transport medium (VTM) using an Omni bead disruptor, clarified at 1500 x g, and at- Store at 80°C until the next embryo passage.

Abstract

Attenuated isolates of viral arthritis / tenosynovitis associated with avian reovirus in poultry are presented, including avian reovirus strains 94826C140 preserved in ATCC with patent name PTA-126077, and progeny or derivatives thereof, as well as avian reovirus strains 96139C140 preserved in ATCC with patent name PTA-126078, and progeny or derivatives thereof. Compositions and methods for administering the isolates or compositions as vaccines to control reovirus-induced viral arthritis / tenosynovitis in galliformes avian are also presented.

Description

[0001] Continue to apply for data [0002] This application claims the benefit of US Provisional Application Serial No. 62 / 942,426, filed December 2, 2019, which is incorporated herein by reference. Background technique [0003] Avian reoviruses are associated with several diseases in poultry, including malabsorption syndrome and growth failure and nutrient malabsorption syndrome (RSS), although these avian reoviruses act as primary sources of these clinical syndromes The role of sexual pathogens is unclear. In contrast, the association of avian reovirus with clinical cases of viral arthritis / tenosynovitis is fairly clear, since reoviruses have been isolated from tendons of affected birds. Control of reovirus-induced viral arthritis / tenosynovitis can be achieved by vaccinating broiler breeders with live and / or inactivated vaccines in combination with maternal immunization delivered to offspring for early protection against field challenge and reach. In broilers, live attenu...
