Embodiment 1

[0059] Aliquot inoculum (A 600 = 1.0) were grown in batch culture (37°C, anaerobic) under constant stirring. Carbohydrate-free batch medium (PYG) was supplemented with glucose (3.0g / L), fructooligosaccharide (FOS, 3.0g / L), sialyllactose (3'- or 6'-sialyllactose, 100mg / L), or fructooligosaccharide plus sialyllactose (3.0g / L and 100mg / L respectively). Growth was measured at various time points by titering the bacteria on selective agar (BIM-25 agar for Bifidobacteria, Wilkens-Chalgren agar for Clostridium and Bacteroides) (see figure 1 ). The results of the growth assay are shown in figure 1 .

[0060] Such as figure 1 As shown, the substitution of 3'-sialyllactose for glucose in the growth medium resulted in substantially no growth of Clostridium or Bacteroides compared to glucose. However, Clostridium and Bifidobacterium infantis, but not Bacteroidetes, grew in the presence of fructooligosaccharides similar to that in the presence of glucose. The combination of fr...

Embodiment 2

[0062] An aliquot inoculum (A 600 = 1.0) were grown in batch culture (37°C, anaerobic) under constant stirring. Carbohydrate-free batch medium (PYG) was supplemented with glucose (3.0g / L), fructooligosaccharide (3.0g / L), or fructooligosaccharide (3.0g / L) plus sialyllactose (3'- or 6'-sialyllactose, 100mg / L). Growth was detected at various time points by measuring the titer of Bifidobacterium lactis on selective agar (BIM-25 agar) (see figure 2 ). The results of the growth assay are shown in figure 2 .

[0063] figure 2 It shows that the growth of Bifidobacterium lactis in the presence of fructooligosaccharides is obviously better than that in the presence of glucose. However, the growth of Bifidobacterium lactis was significantly enhanced in the presence of the combination of fructooligosaccharide and 3'- or 6'-sialyllactose for 46 hours.

Embodiment 3

[0065] A metabolic by-product of bifidobacteria is short-chain fatty acids, the presence of which lowers intestinal pH.

[0066] image 3 Analysis at different times after inoculation of batch cultures of Clostridium, Bacteroides and Bifidobacterium lactis is represented. Aliquot inoculum (A 600 = 1.0) were grown in batch culture (37°C, anaerobic) under constant stirring. Carbohydrate-free batch medium (PYG) was supplemented as described above and the pH of the medium was measured at various time points. The results of the pH analysis are shown in image 3 .

[0067] image 3 As can be seen in , all three colonic bacteria lowered the pH in the presence of glucose. In the presence of sialyllactose alone, the pH increased in all cases. This is consistent with the poorer microbial growth observed in the presence of sialyllactose alone. In the presence of fructooligosaccharides, the pH decreased, but to a lesser extent in batch cultures of Clostridium and Bifidobacterium i...

Abstract

A nutritional composition is provided which comprises oligofructose and sialyllactose.

Description

field of invention [0001] The present invention relates to nutritional formulations containing prebiotic substances and the use of these formulations for promoting the growth of beneficial microorganisms and inhibiting pathogenic organisms. More specifically, the present invention relates to nutritional formulas containing fructooligosaccharides and sialyllactose. Background of the invention [0002] Fructooligosaccharides are a series of natural oligosaccharides found mainly in vegetables, such as onion and chicory plant roots. Oligofructose is considered a specific substrate for bifidobacteria. (See, eg, Mitsuoka et al., "Effect of Fructo-oligosaccharides on Intestinal Microflora", Die Nahrung, 3, 5-6: 427-436 (1987)). [0003] Oligofructose passes through the small intestine without being digested and reaches the large intestine. In the large intestine, fructooligosaccharides are only fermented by a limited range of microorganisms, including most species of bifidobacte...
