Embodiment 1

[0052] Example 1: Take the last 4 to 5 stem nodes and branch nodes of the top of the robust plant of the Xi'an Botanical Garden Chinese Medicine Park as explants, remove the leaves, rinse with running water, dip and shake in 70% ethanol for 30 seconds, and use 0.1% HgCl 2 Disinfect the surface for 9 minutes, rinse with sterile water for 6 times, then cut into 2-leaf sections in sterile water containing 50mg / L Vc, and place them horizontally or vertically on MS medium inorganic salt + B5 medium organic matter + Vc100mg / L+CH300mg / L+6-BA 1.5mg / L+IBA0.2mg / L+AgNO 3 On the medium of 10mg / L+40g / L sucrose, the axillary buds began to germinate and grow after 5 days, and after 25 days, they could grow into aseptic seedlings with a height of 6cm, 6-8 leaves, and 2-4 lateral buds ( figure 1 shown).

[0053] Cut the stem nodes and branches of sterile test-tube plantlets into sections with 2 leaves, and inoculate them in MS medium inorganic salt + B5 medium organic matter + Vc100mg / L+CH300...

Embodiment 2

[0055] Example 2: Take the 40d-year-old robust plant stem nodes and branches of the Xi'an Botanical Garden Chinese Medicine Park as explants, remove the leaves, rinse with running water, dip and shake in 70% ethanol for 30 seconds, and use 0.1% HgCl 2 Disinfect the surface for 8 minutes, rinse 6 times with sterile water, then cut into 2-leaf sections in sterile water containing 150mg / L Vc, and place them horizontally or vertically on MS medium inorganic salt + B5 medium organic matter + Vc100mg / L+CH300mg / L+6-BA1.0mg / L+IBA0.2mg / L+AgNO 3 On the medium of 10mg / L+40g / L sucrose, the axillary buds began to germinate and grow after 5 days, and they could grow into sterile seedlings with a height of 4-6cm, 5-7 leaves and 2 lateral buds in about 30 days.

[0056] Cut the stem nodes and branches of sterile test-tube plantlets into sections with 2 leaves, and inoculate them in MS medium inorganic salt + organic matter in B5 medium + Vc100mg / L+CH300mg / L+6-BA1.5mg / L+ On the culture medium...

Abstract

The invention discloses a method of fast tissue culturing phyllanthus urinaria. The technological character is in that it takes the stem and branch of phyllanthus urinaria as explant, comprising following steps: (1) inducing bud sprout: inoculating the segment of stem and branch of phyllanthus urinaria into bud sprout inducing culture medium MSP1 for germ-free bud sprout differentiation; (2) breeding bud sprout: culturing the segment of stem and branch of said bud sprout on bud sprout breeding culturing medium MSP2, germinating clump sprout which can be used for next breeding process; (3) root growing of tissue culturing sprout: inoculating the single tissue culturing sprout on root growing culturing medium MSP3. The invention provides a new source for explant, which greatly increases breeding radix number. It also shortens the axillary bud generating time through improving culturing medium content and hormone type and proportion content, and inhibits the browning reaction of explant and bud sprout, which greatly increases the breeding coefficient for each explant.

Description

technical field [0001] The invention relates to a method for tissue culture and rapid propagation of Phyllanthus phylloxera, which belongs to the technical field of plantlet cultivation by biological tissue. Background technique [0002] At present, the number of hepatitis B virus carriers is very alarming. According to the statistics of the World Health Organization, there are 400-500 million people in the world, and about 1 million people die from hepatitis B virus infection every year. About 10% of my country's population suffers from this disease, and there is no very effective medicine so far, and the course of treatment is longer. Euphorbiaceae (Phyllanthus urinaria L), as one of the anti-HBV drugs of traditional Chinese medicine, has undergone in-depth research from experiment to clinical in recent years [Zhong Ying, Zuo Chunxu, Li Fengqin, et al. Study on chemical constituents of Phyllanthus phylloxera and its anti-hepatitis B virus activity. Chinese Journal of Tr...
