Embodiment Construction

[0050] 1. Determination of target strains for gene chip detection

[0051] The present invention intends to select 8 kinds of food-borne pathogenic bacteria that are more common in Guangdong Province from the above-mentioned strains for the development of the detection gene chip, and they are respectively Listeria monocytogenes (LM), Vibrio cholerae (VC), Vibrio haemolyticus (VP), Salmonella (Sal), Shigella (Shi), Staphylococcus aureus (SA), Campylobacter jejuni (CJ) and Escherichia coli (EC).

[0052] Sample source: All bacterial strains are from the Guangdong Provincial Center for Disease Control and Prevention.

[0053] The oligonucleotide detection gene chip developed for these 8 kinds of food-borne pathogenic bacteria will be named "Biosafood-8 chip" below.

[0054] 2. Design of oligonucleotide (Oligo) probe

[0055] A total of 351 probes were designed for 89 detection target genes of 8 foodborne pathogens, including 1 positive control probe and 3 negative control probe...

Abstract

The invention relates to a composite gene chip for detecting food pathogenic microbes, which comprises eight parallel sub-arrays constructed by detection probe groups for detecting each microbe in the eight food pathogenic microbes, blank lines are respectively arranged among the eight sub-arrays to be as intervals, thus constructing a big array; the eight food pathogenic microbes are listeria monocytogenes, comma bacillus, vibrio parahaemolyticus, salmonella, shigella, staphylococcus aureus, campylobacter jejuni and escherichia coli. The gene chip can clearly and accurately detect the species of eight target food pathogenic microbes, and also can definitely exclude other off-target microbe samples out of the eight target food pathogenic microbes. Each food pathogenic microbe detection sub-array on the chip can clearly and definitely distinguish off-target microbe samples from target microbe samples.

Description

technical field [0001] The invention belongs to the field of food safety and relates to a composite gene chip for detecting food-borne pathogenic bacteria. Background technique [0002] Food poisoning refers to non-infectious (not infectious) acute and subacute diseases that occur after ingesting food containing biological or chemical toxic and harmful substances or taking toxic and harmful substances as food. According to the pathogenic substances, food poisoning can be divided into five types: bacterial, chemical, animal, plant, and fungal, among which bacterial and chemical food poisoning are the most common. Bacterial food poisoning refers to a disease with acute gastroenteritis as the main symptom of poisoning caused by eating food contaminated by pathogenic bacteria or their toxins. It is the most common type of food poisoning. Common pathogens causing bacterial food poisoning are: Vibrio parahaemolyticus, Salmonella, Staphylococcus, Escherichia coli, Vibrio cholerae,...
