Problems solved by technology

Different from mammals, the liver is not only the main place for the synthesis and secretion of leptin in birds, but also the most important fat metabolism organ in birds. However, whether maternal leptin or leptin treatment in the early stage of avian embryonic development will affect the liver leptin of embryos and newborn chicks Synthesis and secretion, and programmatic effects on hepatic fat metabolism patterns, have not been reported

Embodiment 1

[0048] Three yellow broiler eggs were selected and randomly divided into two groups. The control group was injected with 100 μL solvent (PBS) in the egg white, and the test group was injected with 100 μL containing 0.5 μg mouse recombinant leptin (498-OB-01M, R&D, USA) (Physiol Res 2003 ; 52(2):201-9) PBS. Weigh and take blood samples and liver tissue samples at 12 and 17 embryonic ages and the day of hatching (0 days old) respectively. Serum and liver leptin concentrations were measured by radioimmunoassay, real-time fluorescent RT-PCR was used to quantify liver LEPR and fat metabolism-related genes, and Western blot was used to measure liver LEPR protein content. The results showed that the hatching weight of the leptin treatment group was extremely significantly reduced (P=0.000), while the hepatic body index was significantly increased (P=0.018), especially the difference in females was extremely significant (both hatching weight and hepatic body index were P=0.000 ) (see...

Embodiment 2

[0066] Three-yellow broiler eggs with similar egg weights were selected in this experiment and randomly divided into three groups: the control group (control, Con) was injected with 100 μL of solvent (PBS) in the egg white, and the leptin treatment group was injected with 0.5 μg / 100 μL of PBS (low dose group). , LL) and 5.0 μg / 100 μL PBS (high-dose group, HL) mouse recombinant leptin, liver and serum samples were collected after hatching until 21 days old. The concentrations of leptin and thyroid hormone in liver and serum were determined by radioimmunoassay, the content of LEPR protein in chick liver was determined by Western blot, and the expressions of liver LEPR and fat metabolism-related genes were quantified by real-time fluorescent RT-PCR. The results showed that in ovo injection of leptin affected the early growth of broiler chickens after hatching in a sex-specific and dose-dependent manner (see Table 2-1, 2-2, 2-3). Regardless of low dose or high dose, the chick weig...

Abstract

The invention discloses a method for injecting leptin into egg white, and belongs to the technical field of animal growth regulation. The method comprises: before incubation, disinfecting the small end of a hatching egg; drilling a hole lightly on the disinfected part by a toad marrow puncture needle; injecting leptin injection reagent into the egg white through the hole by a micro injector; and sealing the hole by paraffin. After the leptin with certain concentration is injected into the egg white, the leptin is observed to inhibit the development of embryo of table poultry and reduce body weight of newly hatched offspring, but promote early growing development of the table poultry after hatch, and the leptin can be taken as semiochemicals to mediate the routinization effect on the early growing development of filial generations of the table poultry.

Description

1. Technical field [0001] The invention relates to a method for injecting leptin into egg white. Specifically, the invention relates to a method of using leptin as a signal substance to mediate the programmed effect on the early growth and development of broiler offspring, and belongs to the technical field of animal growth regulation. 2. Background technology [0002] In the 21st century, obesity has become an important public health problem and has attracted more and more attention from the world. With the development of economy and the improvement of material life in our country, more and more children have quietly joined the ranks of obese people. Obese children have accounted for 10% of the total number of children, and are increasing at an annual rate of 8%. Today, the requirements for animal food quality are increasing day by day, because the lipid deposition of meat livestock and poultry is directly related to the quality of meat products, the problem of animal obesi...
