Problems solved by technology

[0005] It can be predicted that the design and preparation of HBV infection-blocking cell-entry inhibitory polypeptides represents a new anti-HBV treatment strategy, which is one of the internationally competitive and challenging topics. At present, there is no relevant research work reported in China.

Embodiment Construction

[0020] The object of the present invention can be achieved through the following measures:

[0021] 2. The present invention selects the 2-48, 2-38 and 2-28 amino acids of the pre-S1 region of the hepatitis B virus B genotype envelope protein to design small molecular polypeptides, which are characterized in that they contain the following three amino acid sequences: A kind of:

Abstract

The invention discloses a novel small-molecule polypeptide blocking hepatitis B virus infection, belongs to the technical field of genetic engineering, designs an amino acid sequence polypeptide of 2-48 position, 2-38 position and 2-28 position of a front S1 area of a hepatitis B virus B gene type envelope protein according to the characteristics of an amino acid sequence of the B type HBV envelope protein, and studies the treatment effect of the polypeptide in blocking the virus infection and verifies the validity and the feasibility of the polypeptide in an HBV infected HepRG cell model and a marmot hepatic cell model. The invention comprises the following steps: selecting, modifying and changing three small-molecule polypeptide amino acid sequences; comparing the difference between the antiviral treatment effects of polypeptides with different modifications, different lengths and different gene mutations; selecting a high-efficiency and optimized viropexis inhibition molecule; and determining an optimal medicament dosage. The invention also systematically studies the effect of the polypeptide in blocking the HBV virus infection in HBV infection models in vitro and in vivo and lays a good experimental foundation for further antiviral study in vivo.

Description

Technical field: [0001] The invention belongs to the technical field of genetic engineering. Background technique: [0002] Hepatitis B virus (Hepatitis B virus, HBV) infection is one of current serious global health problems, there are about 400,000,000 people in the world infected with HBV, mainly concentrated in developing countries; and my country is a high prevalence of hepatitis B virus infection In China, there are about 120 million chronic hepatitis B virus carriers, a considerable number of infected persons (about 30 million cases) develop into chronic hepatitis, and a few also develop into liver cirrhosis. There are about 1 million deaths from hepatitis B-related diseases (mainly including liver cirrhosis and liver cancer) every year in the world, and about 300,000 cases in my country, accounting for the first cause of death among all infectious diseases. Hepatitis B is therefore a public health and social problem that needs to be addressed with priority. [0003]...
