[0026] 1.1 Materials

[0027]Petunia hybrida (Petunia hybrida) variety is QL01, and sterile seedlings were obtained by tissue culture of isolated leaves. The wild-type Agrobacterium rhizogenes K599 was preserved by the Key Laboratory of Botany, School of Life and Environmental Sciences, Hangzhou Normal University, and the plasmid pSLD50-6 containing the CYP2E1 gene (see figure 1 ) and the control plasmid pKH200 containing the GUS gene were donated by Professor Doty of Washington University.

[0029] Plasmids pSLD50-6 and pKH200 were transformed into Agrobacterium rhizogenes K599 by freeze-thaw method, that is, about 0.1 μg of purified plasmid DNA was added to 100 μL of K599 competent cells, mixed well, placed on ice for 10 minutes, and placed in liquid nitrogen for quick freezing for 5 minutes , Immediately heat-shock in a 28°C water bath ...

Abstract

The invention provides a cultivating method of transgenic petunia capable of removing environmental pollutants efficiently, which comprises the following steps: a vector pSLD50-6 containing a CYP2E1 gene is transferred to agrobacterium rhizogenes K599; transgenic hairy adventitious roots transformed by the CYP2E1 gene are obtained by an agrobacterium tumefaciens transgenic method; the hairy adventitious roots induce callus on a culture medium of MS + BA 1.0mg/L; the callus obtains cluster buds on a culture medium of MS + BA 1.0mg/L + NAA 0.4mg/L; and the cluster buds regenerate plants on a culture medium of MS + NAA 0.1mg/L to obtain the transgenic petunia. The petunia obtained in the invention can be used for removing indoor air pollutants of benzene, toluene and the like, and is very favorable to the physical and mental health of people, the improvement of the quality of life of people, and the construction of an ecological environment.

Description

(1) Technical field [0001] The invention relates to a method for cultivating transgenic petunias that can efficiently remove environmental pollutants such as benzene and toluene. (2) Background technology [0002] Benzene and toluene are widely used in chemical raw materials such as paints, dyes, adhesives, wallpapers, carpets, synthetic fibers, cleaning agents and some organic solvents, and are one of the main harmful gases for indoor environmental pollution; Machines, computer terminals and printers also produce benzene and toluene pollution during the use process, tobacco smoke and gasoline exhaust also contain benzene and toluene, and benzene and toluene are extremely carcinogenic (Wang Guihua, Zhou Zhongping, Fu Lixin. "Investigation on the Distribution Characteristics of Benzene and Toluene Concentration in Indoor Air". Shanghai Environmental Science, 2003, 22(12): 977-978, 982; Shen Xueyou, Luo Xiaolu, Zhu Lizhong. "Research Progress of Volatile Organic Compounds in A...
