The handheld bioluminescent detector is characterized by comprising an optical detection module 1, a signal acquisition module 2, a signal processing module 3, a signal display and transmission module 4 and a power supply module 5. The detection method for the handheld bioluminescent detector is characterized by comprising the steps: converting bioluminescent signals to be detected into electric signals by the optical detection module 1, transmitting the electric signals to the signal processing module 3 by the signal acquisition module 2, displaying and transmitting the completed result by the signal processing module, and simultaneously providing feedback control quantity to the optical detection module 1 by the signal processing module. The power supply module 5 is used for supplying a proper working voltage to all the modules. By means of the detector and the detection method provided by the invention, the traditional problem that weak bioluminescent signals are difficult to detect by using a handheld instrument is solved effectively.
