Problems solved by technology

[0003] However, in the interspecific hybridization of wild chrysanthemum and cultivated chrysanthemum, the problem of low seed setting rate and even inability to obtain hybrid seeds is often encountered, which makes it difficult to introduce many excellent genes of low ploidy wild chrysanthemum into cultivated varieties to achieve their resistance improvement

Embodiment 1

[0022] 1. Use the commercially available cultivated chrysanthemum 'Yuhua Xingchen' as the female parent, select well-developed flower buds, emasculate when the tongue-shaped flowers are just exposed, and remove all the bisexual tubular florets in the inner ring, and cut off the tongue-shaped flower petals at the same time Until the stigma is visible (the stigma cannot be hurt), and bagged strictly. The male parent is isolated by bagging before the flower heads open and loose powder. When the stigma of the female parent protrudes and splits to present acute angles and secretes mucus (3 to 5 days after emasculation), collect fresh pollen from the bagged chrysanthemum brain of the male parent, pollinate the female parent with a brush, and pollinate the female parent immediately after pollination. Ben continued to be bagged and quarantined. 1h and 24h after pollination, the pollinated inflorescences were cut respectively, immediately snap-frozen in liquid nitrogen and stored at -...

Abstract

The invention discloses a method for quickly identifying differential protein during interaction of chrysanthemum pollen and stigma, belonging to the field of chrysanthemum proteomics. The method comprises the following steps of: performing distant hybridization work of chrysanthemum by taking cultured chrysanthemum as a female parent and wild chrysanthemum as a male parent; cutting off pollinated inflorescences in 1 h and 24 h after pollination; removing petals and sepals of ligulate flowers in the obtained inflorescences to obtain pistils; extracting a protein sample from the obtained pistils by using a TCA (Trichloroacetic Acid) / acetone precipitation method; and quickly identifying the differential protein by applying iTRAQ (Isobaric Tags For Relative And Absolute Quantitation) and a mass spectrum technology and performing bioinformatic analysis. Aiming at the chrysanthemum and wild species of related genera thereof, the invention provides a method for quickly and accurately identifying key differential protein during the interaction of the chrysanthemum pollen and the stigma for providing research foundation for solving the problem of distant hybridization incompatibility of the chrysanthemum.

Description

technical field [0001] The invention relates to the field of proteomics of chrysanthemums, in particular to a rapid detection and identification method for differential proteins in the interaction between pollen and stigma after hybridization of chrysanthemums. Background technique [0002] Chrysanthemum is one of China's top ten traditional famous flowers and one of the world's four major cut flowers. It has a wide variety, rich colors, changeable flower shapes, and various postures. It plays an extremely important role in the cultivation and application of flowers in the world. Chrysanthemum is native to China and has a cultivation history of more than 1,600 years. At present, there are more than 4,000 chrysanthemum varieties selected and bred in China, and the total number of varieties in the world exceeds 20,000. However, most chrysanthemum varieties have poor stress resistance, which seriously limits their Yield and quality improvement and promotion application geograph...
