Problems solved by technology

[0003] The deficiency that above method exists, the one, utilize mature wheat seed to carry out generation, need to sow mature seed, this is compared with utilizing immature embryo one-step seedling, the time from pollination to immature embryo maturity is longer; The 2nd, embryo seedling is transplanted in Greenhouse growth requires ventilation and cooling to the greenhouse in summer, and the cost of scientific research is relatively high; the second is to choose wheat with strong spring nature, which is not conducive to popularization and application

Embodiment 1

[0019] Example 1 First-year winter wheat immature embryo one-step seedling transplantation in summer field rapid growth experiment

[0020] 1. Planting of parent material

[0021] On October 6, 2008, the mature seeds of winter wheat Jimai 20 and Yanyou 361 were sown in the agronomy experimental field of Shandong Agricultural University.

[0022] 2. Get contemporary hybrid F 1 immature embryo

[0023] Using the conventional sexual hybridization method, Jimai 20 was emasculated for 3-5 days, and on May 1, 2009, the mature pollen of Yanyou 361 was smeared on the stigma of the female parent Shannong 15 to obtain the contemporary hybrid F 1 Immature embryos, namely Jimai 20×Yanyou 361→F 1 Immature embryos were placed, and the date of pollination was marked on the hybrid ears.

[0024] 3. Young embryos grow into seedlings in one step: on May 15, 2009, the above-mentioned contemporary hybrid F 1 Immature embryos on day 14 were sterilized with 75% ethanol for 8 minutes, rinsed wi...

Embodiment 2

[0031] Example 2 One-step seedling transplantation of winter wheat immature embryos in the second year to field field rapid growth test in summer

[0032] 1. Planting of parent material

[0033] On October 7, 2009, the winter wheats Tainong 18 and Zhongyou 9507 were sown in the agronomy experimental field of Shandong Agricultural University.

[0034] 2. Get contemporary hybrid F 1 immature embryo

[0035] On May 2, 2010, using conventional sexual crossing method, obtained Tainong 18×Zhongyou 9507→F 1 contemporary hybrid immature embryos,

[0036] Thereafter from F 1 Embryo inoculation, vernalization of embryo seedlings, site fertilization, hardening, transplanting of embryo seedlings, field management of embryo seedlings, etc. are the same as the one-step seedling transplantation of first-year winter wheat embryos in the summer field in Example 1. The difference is that The embryonic age of inoculated immature embryos was 16 days, and the vernalization time was 20 days. ...

Embodiment 3

[0037] Example 3 One-step seedling transplantation of winter wheat immature embryos in the third year to field field rapid growth test in summer

[0038] 1. On October 5, 2010, the winter wheat Jimai 20 and Nongda Youfan 5 were sown in the agronomy experimental field of Shandong Agricultural University.

[0039] 2. Get contemporary hybrid F 1 immature embryo

[0040] On May 5, 2011, Jimai 20 × Nongda Youfan 5 → F was obtained by conventional sexual hybridization 1 contemporary hybrid immature embryos,

[0041] Thereafter from F 1 The method from inoculation to transplanting was the same as the one-step seedling transplantation of winter wheat immature embryos in the previous two years in the summer field plus generation experiment, the difference was that the immature embryos were inoculated for 14 days and vernalized for 20 days.

[0042] The focus of the present invention is to first inoculate the 14-16d immature embryos of winter wheat on the MB medium to obtain one-step ...

Abstract

The invention discloses a novel method for transplanting winter wheat young embryo one-step seedlings big summer fields for fast growing. According to the method, a young embryo one-step seedling formation technology, a technology for transplanting embryo seedling in the big summer fields for growth and so on are adopted, winter wheat can grow fast in the big summer fields by utilizing natural sunlight. The method comprises the following steps: inoculating 14-16 d young embryos in an MB culture medium, wherein the emergence rate and embryo seedling transplanting survival rate are higher; placing the embryo seedlings in an illumination culture box at 0-6 DEG C for an appropriate time of 20-25 d; transplanting the embryo seedlings to backs of big field furrows in middle June, so that flowering periods are kept away from summer periods, wherein the embryo seedling survival rate and the maturing rate are increased by using embryo seedling transplanting management measures; and scientific research input is reduced because the seedlings fast grow by using natural sunlight.

Description

technical field [0001] The invention relates to a new method for one-step seedling transplantation of immature winter wheat embryos to a summer field for rapid cultivation, belonging to the field of crop breeding. Background technique [0002] Wheat breeding experts at home and abroad are making unremitting explorations in order to shorten the wheat breeding cycle and speed up the wheat breeding process. Wang Hengli (1987), Guo Wenxiu (1993), Mi Guohua et al. (1998), Yao Jinbao et al. (1998) took advantage of the special geographical environment with large differences in latitude or altitude and the climatic conditions in different regions to plant mature wheat seeds and carry out Wheat breeding. Wang Zhiwei et al. (1999) selected wheat with strong spring nature to achieve rapid development of wheat. Ding Xiaoyi et al. (2005) adopted the technology of in vitro culture of immature embryos, and transplanted wheat immature embryo seedlings in the greenhouse for additional gen...
