[0023] A total of 51 clinical samples of osteosarcoma were selected, fixed with 10% formaldehyde solution for 7 days, and then paraffin-embedded tissue blocks were prepared. A paraffin section with a thickness of 8 microns was prepared from the sample, and the expression of DJ-1 protein (the amino acid sequence is shown in SEQ ID No: 1) was detected by immunohistochemical method, and at the same time, another section was selected for H&E staining. The expression of DJ-1 protein and H&E staining were observed under an upright microscope. see figure 1 , the expression of DJ-1 protein in the sample cancer tissue was higher than that in the non-tumor tissue in the same film.

After 24 hours of transfection, cisplatin, paclitaxel, doxorubicin and methotrexate were administered respectively, and the growth inhibitory effect of the drugs on the cells was detected. see Figure 5 with Image 6 , KHOS and U2OS osteosarcoma cells significantly increased drug sensitivity after knocking out the DJ-1 gene.
