Embodiment 1

[0019] Broccoli seeds were used as raw materials. During the germination process, they were heat-shocked once a day at 40°C for 20 minutes each time. When the seeds germinated to the 10th day, the sprouts were harvested, and the ratio of solid to liquid was 1:5 ( w / v) Grind the sprouts into a homogenate with distilled water. According to known techniques, the homogenate was incubated at 37°C for 3 hours to hydrolyze endogenous glucosinolates into isothiocyanates, and then extract the isothiocyanates with dichloromethane, and centrifuge to obtain isothiocyanate-rich The extract of salt, its content is 5 times of before treatment.

Embodiment 2

[0021] Taking cabbage seeds as raw materials, during the germination process, place them at 50°C for heat shock for 15 minutes every day, and harvest the sprouts when they germinate to the 8th day. ) Homogenize the sprouts with distilled water. Incubate, extract, and centrifuge according to the method of Example 1 to obtain an extract rich in isothiocyanate, the content of which is 7 times that of before treatment.

Embodiment 3

[0023] Using kale seeds as raw materials, heat-shock them at 60°C for 15 minutes during the germination process. When they germinate to the 8th day, harvest the sprouts and use them according to the ratio of material to liquid 1:5 (w / v). Homogenize the sprouts with distilled water. Incubate, extract, and centrifuge according to the method of Example 1 to obtain an extract rich in isothiocyanate, the content of which is 10 times that of before treatment.

Abstract

The invention relates to a high-efficiency enrichment technology for isothiocyanate in brassica bud seedlings. The technology is characterized by comprising the following steps: in 5 to 10 d after germination of brassica seeds, subjecting the seeds to heat shock at 40 to 70 DEG C for 10 to 20 min once each day; after harvesting of bud seedlings, grinding and homogenizing the harvested bud seedlings with distilled water according to a material-liquid ratio of 1: 5-10 (w / v), carrying out incubation at 37 DEG C for 3 h and allowing endogenous glucosinolate (thioglycoside) to be thoroughly hydrolyzed so as to produce isothiocyanate with an anticancer effect; and extracting isothiocyanate in a hydrolysate with dichloromethane and carrying out centrifugation so as to prepare an extract rich in isothiocyanate. The technology has the advantages of a wide application scope, simple operation and low production cost. After the brassica bud seedlings have undergone heat shock and homogenization, the content of produced isothiocyanate is 5 to 10 times of the content of isothiocyanate before treatment, and high-efficiency enrichment of isothiocyanate is realized.

Description

1. Technical field [0001] The invention relates to a high-efficiency enrichment technology of isothiocyanate in sprouts of Brassica plants, especially the enrichment of isothiocyanate in sprouts of plants such as cabbage, broccoli and cauliflower, and belongs to the field of food biotechnology. 2. Background technology [0002] Isothiocyanate is currently recognized as a plant active substance with great anticancer activity, which can significantly inhibit type I enzymes and activate type II detoxification enzymes, thereby playing anticancer and antitumor effects. Brassica sprouts are rich in aliphatic glucosinolates, and the content of glucosinolates in seedlings is significantly higher than that in mature tissues. When plant tissue is destroyed, glucosinolates are hydrolyzed by contact with myrosinase to produce isothiocyanate, so Brassica plant sprouts are a good source of isothiocyanate. However, most Brassica plants contain ESP protein, which inhibits the hydrolysis of...
