Embodiment 1

[0021] Example 1: Raw material 2.0 g of commercially available pectin was weighed, dissolved in 100 mL of distilled water, dissolved and swelled completely at room temperature, and sterilized under ultraviolet light for 10 h. Add 100 mL of 0.1 mol / L NaOH solution into the same container, stir well, make the pectin concentration 1wt%, NaOH concentration 0.05 mol / L, raise the temperature to 30 ℃ for 2 h, and then use 3 mol / L HCl Adjusted to pH=3, reacted at room temperature for 22 h, the product was desalted by dialysis bag dialysis (molecular weight cut-off 3500 Da), concentrated, and freeze-dried to obtain modified citrus pectin MCP-1. The weight average molecular weight of the product is detected by high performance liquid chromatography M w is 208784 Da.

Embodiment 2

[0022] Example 2: Raw material 2.0 g of commercially available pectin was weighed, dissolved in 100 mL of distilled water, dissolved and swelled completely at room temperature, and sterilized under ultraviolet light for 10 h. Add 100 mL of 0.1 mol / L NaOH solution into the same container, stir evenly, make the pectin concentration 1wt%, NaOH concentration 0.05 mol / L, increase the temperature to 50 ℃ for 4 h, and then use 3 mol / L HCl Adjusted to pH=3, reacted at room temperature for 22 h, the product was desalted by dialysis bag (molecular weight cut-off: 3500 Da), concentrated, and freeze-dried to obtain modified citrus pectin MCP-2. The weight average molecular weight of the product is detected by high performance liquid chromatography M w is 103569 Da.

Embodiment 3

[0023] Example 3: Raw material 2.0 g of commercially available pectin was weighed, dissolved in 100 mL of distilled water, dissolved and swelled completely at room temperature, and sterilized under ultraviolet light for 10 h. Add 100 mL of 0.02 mol / L NaOH solution into the same container, stir well, make the pectin concentration 1wt%, NaOH concentration 0.01 mol / L, raise the temperature to 70 ℃ for 1 h, and then use 3 mol / L HCl Adjusted to pH=3, reacted at room temperature for 22 h, the product was desalted by dialysis bag dialysis (molecular weight cut-off 3500 Da), concentrated, and freeze-dried to obtain modified citrus pectin MCP-3. The weight average molecular weight of the product is detected by high performance liquid chromatography M w is 34891 Da.

Abstract

The invention relates to a preparation process and application of MCP (Modified Pectin), and particularly relates to a preparation process of a degradable pectin with different molecular weight ranges and application of the degradable pectin in the anti-tumor field. The preparation process of the MCP comprises the following steps: dissolving and swelling a pectin by using water; sterilizing for 10 hours under an ultraviolet lamp; regulating a solution to be alkaline (the NaOH concentration is 0.01-0.05 mol / L) by using a NaOH solution; degrading at certain temperature (30-70 DEG C) for a period of time (1-4 hours), and then regulating by using 3 mol / L HCl until the pH is equal to 3; reacting at room temperature for 22 hours; and carrying out membrane dialysis, desalination, concentration and freeze drying on a product to obtain the MCP. The preparation process disclosed by the invention is favorable to obtaining the MCP with different molecular weight distributions. The MCP has pancreatic cancer resistant activity and is capable of preferably inhibiting the proliferation of a tumor cell and higher in medical value.

Description

technical field [0001] The invention relates to a preparation process and application of modified pectin, in particular to a preparation process of modified pectin in different molecular weight ranges and its application in the field of antitumor. Background technique [0002] Pectin is an acidic polysaccharide polymer compound extracted from plants. It has the characteristics of natural, green, nutritious and healthy. It has been jointly confirmed by the Joint Expert Committee of the Food and Agriculture Organization of the United Nations and the World Health Organization (FAO / WHO) Food ingredients without ADI (Admissible Daily Intake) limits. At present, pectin has been widely used in food, cosmetics, chemical industry, textile, medicine and other fields, and has played an important role in improving people's life. In the food and pharmaceutical industries, pectin has the functions of antibacterial, hemostasis, detoxification, anti-radiation, lowering blood sugar, prevent...
