Abstract
The invention provides a method for stimulation-induced rooting of crataegus cuneata test-tube plantlets using high concentration auxin. The method comprises the specific steps that crataegus cuneata test-tube plantlets are selected form the sterile crataegus cuneata seedlings which are continuously subjected to multiplication culture of multiple shoots 5 times, and 2-3 pieces of base leaves of the test-tube plantlets are cut; then the crataegus cuneata test-tube plantlets of which the 2-3 pieces of the base leaves are cut are inoculated into 3 mg / L-5 mg / L of IBA auxin solution for stimulation, and the test-tube plantlets are inoculated in a blank culture medium after the stimulation is performed for 1-3 days, then the crataegus cuneata test-tube plantlets are treated in the IBA auxin solution for 1 day, thus the induced rooting culture of the crataegus cuneata test-tube plantlets is completed; the induced rooting seedlings of the crataegus cuneata are transplanted to a mixed soil comprising peat soil and vermiculite, and then the stimulation-induced rooting of the crataegus cuneata test-tube plantlets using the high concentration auxin is completed. The number of the obtained primary roots reaches 4.13 on average, the surviving rate is counted and reaches 88.6% after the hardening-seedlings are transplanted for 3 weeks.
