Problems solved by technology

[0002] At present, most Phalaenopsis tissue culture producers in China use sodium hypochlorite solution or mercuric chloride solution to disinfect the phalaenopsis buds. The mercuric chloride solution has a very good disinfection effect on the pedicel buds, but it contains harmful substances to the environment and soil. Seriously polluting mercury, so many responsible manufacturers have abandoned it
Sodium hypochlorite solution has a good disinfection effect, is easy to clean, and has no pollution. It is the main disinfectant used by domestic Phalaenopsis tissue culture manufacturers. At present, the pollution rate of sodium hypochlorite for disinfection of Phalaenopsis pedicel buds is about 20%. Sodium hypochlorite is a chemical reagent. Buy it Need to go to a special chemical reagent store and the price is higher

Embodiment 1

[0024] Embodiment 1, a kind of disinfection method of Phalaenopsis pedicel bud, its steps are as follows:

[0025] (1) Pedicel selection: select at least three Phalaenopsis plants that bloom in the greenhouse, have no virus manifestations, normal inflorescences, and complete pedicel buds, cut them off at 10 cm from the base of the pedicel with a sterilized tool such as a knife or scissors, and Cut off the upper inflorescence, and only keep the part of the peduncle with the peduncle bud;

[0026] (2) Coarse disinfection of pedicel: Wipe the pedicel with 70%-75% alcohol cotton from bottom to top for 3-5 times; after wiping, wrap it with plastic wrap, write the code and date, and send it to the rear process.

[0027] (3) Grading of pedicel buds: cut off the pedicel buds respectively on the aseptic operating table, and grade them according to the degree of tenderness, and divide them into two types: old pedicel and young pedicel; Cut at a distance of 3cm below the pedicel bud; ...

Embodiment 2

[0034] Embodiment 2, in the disinfection method of the Phalaenopsis pedicel bud described in Example 1, the pedicel bud induction medium is Huabao No. 1 2g / L+coconut juice 50ml / L+6-BA 3PPM+NAA 0.2PPM.

Embodiment 3

[0035] Embodiment 3, in the disinfection method of Phalaenopsis pedicel buds described in Example 1, the pedicel bud induction medium is Huabao No. 1 3.5g / L+coconut juice 100ml / L+6-BA 5PPM+NAA0.5PPM.

Abstract

The invention provides a disinfection method for pedicel buds of phalaenopsis amabilis, which comprises the steps of: 1) selecting phalaenopsis amabilis plants which have complete pedicel buds and remains pedicels with the pedicel buds; 2) wiping the pedicels with alcohol cotton balls for 3-5 times from bottom to top; 3) according to the aged degree of the pedicels, classifying the pedicels into aged pedicels and tender pedicels; 4) preparing three disinfectants, respectively placing the aged pedicels and tender pedicels into two empty sterile bottles, pouring 1# disinfectant into the bottles with the aged pedicels and the 2# disinfectant into the bottles with the tender pedicels, and then pouring the disinfectants out; 5) stripping off the bracts on the pedicel buds on the aged pedicels and the tender pedicels after the first disinfection, soaking the pedicel buds, after the bracts are stripped off, in the 3# disinfectant, and pouring the disinfectant out; 6) inoculating the pedicel buds in a culture bottle filled with a pedicel bud inductive culture medium; and 7) placing the culture bottle in a culture room and performing induction to the pedicel buds. The disinfection method is 2-7% in pollution ratio of the pedicel buds, is 95-99% in survival rate of the pedicel buds, and is low in cost of the disinfectants.

Description

technical field [0001] The invention relates to a method for cultivating and disinfecting pedicel buds, in particular to a method for disinfecting phalaenopsis pedicel buds.
