Embodiment Construction

[0014] In order to make the object, technical solution and advantages of the present invention more clear, the present invention will be further described in detail below in conjunction with the examples. It should be understood that the specific embodiments described here are only used to explain the present invention, not to limit the present invention.

[0015] The application principle of the present invention will be further described below in conjunction with the accompanying drawings and specific embodiments.

[0016] The lipopolysaccharide D-galactosamine-induced liver injury protection animal model provided by the embodiments of the present invention is a male BALB / c mouse (20-25g); and food feeding; the acute liver injury model was established by intraperitoneal injection of LPS (50 μg / kg) and D-GalN (800 mg / kg). Two mice were randomly divided into 6 groups: control group, mangiferin (20mg / kg) group, LPS / GalN group and mangiferin (5, 10 and 20mg / kg)+LPS / GalN group. ...

Abstract

The invention belongs to the field of a medical medicine, and discloses a lipopolysaccharide D galactosamine-induced liver injury animal protection model and a construction method thereof. The mangiferin obviously inhibits generation of LPS / GalN-induced ALT and AST, and at the same time, the histology analysis hepatic tissue shows that the mangiferin obviously inhibits LPS / GalN-induced liver injury. The result shows that mangiferin has protection effect for LPS / GalN-induced liver injury. The mangiferin obviously inhibits generation of LPS / GalN-induced TNF-alpha, IL-1beta, iNOS, MCP-1 and RANTES. By inhibiting oxidative stress, cell apoptosis and inflammation reaction, the mangiferin can protect the LPS / GalN-induced liver injury, and the mangiferin can prevent a LPS / GalN-induced liver injury by activating Nrf2 / HO-1 signal pathway and inhibiting the activation of NLRP3 inflammasome.

Description

technical field [0001] The invention belongs to the field of medical medicine, and in particular relates to a lipopolysaccharide D-galactosamine-induced liver injury protection animal model and a construction method. Background technique [0002] Acute liver injury is a life-threatening syndrome caused by hemorrhagic necrosis and hepatocyte apoptosis, a devastating syndrome associated with high mortality. It has been reported that LPS, a membrane component of Gram-negative bacteria, plays a key role in the process of initiating endotoxin. GalN was able to amplify LPS-induced liver injury within hours. LPS and GalN-induced acute liver injury in mice is similar to clinical acute liver injury and has been used as a promising experimental model. LPS and GalN can induce various inflammatory cytokines, such as IL-1β and TNF-α, which can lead to hepatocyte apoptosis and liver injury. Apart from liver transplantation, there is currently no effective prevention and treatment. The...
