Embodiment 1

[0096] [Example 1] Establishment of L02 stable transfection cell line with TRIM32 overexpression

[0097] According to the steps for establishing the L02 stably-transformed cell line in the embodiment, a L02 stably-transformed cell line with TRIM32 overexpression was established. Afterwards, the cells were collected, and the expression of TRIM32 was verified by WB. The result is as figure 1As shown, it can be seen that in L02 cells infected with the TRIM32 overexpression lentiviral system, the expression of TRIM32 was significantly increased, indicating that the cell line was successfully established.

Embodiment 2

[0098] [Example 2] Effect of TRIM32 Gene Overexpression on Fat Accumulation in Hepatocytes

[0099] 1. Grouping of experimental cells: L02 empty control group, TRIM32 overexpressed L02 stable cell line control group, L02-empty experimental group, TRIM32 overexpressed L02 stable cell line experimental group.

[0100] 3. Establishment and detection of fatty liver cell model: after the cells adhere to the wall and the cells are cultured to 50% healing, add palmitate (PA) and oleic acid (OA) (PA 0.2mM+OA0 .4mM) for stimulation, and the same amount of BSA was added to the control group. After 12 hours, the cell samples of each group were collected for Oil Red O staining experiment.

[0101] The results of Oil Red O staining were as follows: figure 2 As shown, the cells in the control group had no obvious red color, but after adding PA+OA stimulation for 12 hours, the number of cells stained red by Oil Red O increased significantly compared with the control group. Under the same...

Abstract

The invention discloses application of E3 ubiquitinligating enzyme TRIM32 for preparing drugs for treating fatty liver and related diseases. A human normal liver cell L01 cell line and a TRIM32 overexpression L02 cell line are adopted as experimental subjects, and the function of the TRIM32 gene is researched through a fatty liver cell model inducted by palmitic acid and oleic acid. A result showsthat under the stimulus of palmitic acid and oleic acid, the area of red fat droplets of TRIM32 overexpression L02 cells is remarkably increased compared with a control group, and it indicates that TRIM32 overexpression can worsen lipid accumulation generated by combined induction of palmitic acid and oleic acid; in other words, the TRIM2 gene can promote development and progression of the fattyliver and related diseases. Accordingly, TRIM32 provides a target for preparing drugs for preventing, relieving and / or treating the fatty liver and related diseases.

Description

technical field [0001] The invention belongs to the field of gene function and application, and relates to the function and application of E3 ubiquitin ligase TRIM32 in the treatment of fatty liver, and the application of TRIM32 as a target gene in the development of drugs for preventing, alleviating and / or treating fatty liver disease. Background technique [0002] Nonalcoholic fatty liver disease (NAFLD) is the most common liver disease in the world, with an incidence rate of 15% to 30% in Western countries, 27% in Hong Kong, China, and 27% in Southeast Asia, South Korea, Japan and Taiwan. 15% to 45%, and 20% in mainland China[1]. Nonalcoholic fatty liver is divided into two categories, one is the milder nonalcoholic fatty liver (NAFL), and the other is the more severe nonalcoholic steatohepatitis (NASH). The continued development of non-alcoholic fatty liver can lead to cirrhosis and liver cancer, which seriously threatens human health [2]. [0003] The liver is the cen...
