Embodiment Construction

[0040] The inventor collected a large number of venous serum and plasma samples from the First Affiliated Hospital of Nanjing Medical University from 2014 to 2016, and selected 290 cases of breast cancer and 402 cases of breast cancer by sorting out the sample data. The normal control sample was used as the experimental sample for qRT-PCR verification. At the same time, 32 cases of breast cancer tissues and 32 cases of paracancerous tissues were retained. The selected serum samples of patients were all from newly diagnosed patients who had not undergone surgery, radiotherapy and chemotherapy intervention and were pathologically confirmed as breast cancer. The demographic data and clinical data of these samples were collected systematically.

[0041] Refer to the flowchart ( figure 1 ), for the 32 miRNAs from three clusters located on the X chromosome, through the training set and the verification set, the relative quantitative method based on qRT-PCR is used for verification...

Abstract

The invention discloses a circulating miRNA (micro Ribonucleic Acid) marker related to breast cancer auxiliary diagnosis and located on an X chromosome and application thereof. The marker is one or more of blood plasma markers miR-106a-3p, miR-106a-5p, miR-20b-5p, miR-92a-2-5p, miR-500a-5p, miR-501-5p, miR-188-3p, miR-let-7f-5p and miR-98-5p, and blood serum markers miR-106a-5p, miR-19b-3p, miR-20b-5p, miR-92a-2-3p, miR-501-3Rp, miR-502-3p, miR-532-3p, miR-532-5p and miR-188-3p. Circulating miRNA is used as a novel biological marker and has the advantages of good stability, minimal invasive and easiness for obtaining and high sensitivity and specificity. The development and utilization of the biological marker provides a new direction for diagnosis of various diseases including tumors andfurther treatment. The research provided by the invention more pertinently obtains the breast cancer circulating miRNA marker with a clinical diagnosis potential. The research proves that the reliability and repeatability of taking the miRNA as a noninvasive marker for diagnosing the breast cancer.

Description

technical field [0001] The invention belongs to the fields of genetic engineering and oncology, and relates to a circulating miRNA marker located on X chromosome related to auxiliary diagnosis of breast cancer and application thereof. Background technique [0002] Breast cancer (Breast Cancer, BC) is the most common malignant tumor in women. According to the latest World Cancer Report (World Cancer Report 2014), it accounts for 25.2% of malignant tumors in women worldwide. In China, the incidence of breast cancer is on the rise. In 2015, the incidence of breast cancer in Chinese women ranked first among malignant tumors, and the mortality rate ranked sixth. Existing examination methods such as breast ultrasonography, mammography, nuclear magnetic resonance and hollow needle biopsy all require a certain volume of tumor tissue to detect, and there are certain limitations in the early diagnosis of breast cancer; and clinical Commonly used tumor markers such as carbohydrate ant...
