Embodiment 1

[0035] 1. Experimental animal samples

[0036] 53 SPF grade Wistar rats, weighing 170-240g, 1.5 months old; 53 SPF grade GK rats, weighing 150-220g, 1.5 months old. All were purchased from Shanghai Slack Experimental Animal Co., Ltd. [SCXK (Shanghai) 2017-0005]. 50 SPF grade BN rats, weighing 70-150 g, aged 1 month; 50 SPF grade SD rats, weighing 80-150 g, aged 1 month. All were purchased from Beijing Weitong Lihua Experimental Animal Technology Co., Ltd. [SCXK (Beijing) 2016-0011]. The rat tail tissues of rats were collected from the animal experiment facilities of the Guangdong Provincial Laboratory Animal Monitoring Institute [SYXK (Guangdong) 2016-0122], and the ethics review number was IACUC2014029.

[0037] 2. The initial screening process of SNP

[0038] First, the DNA of 3 Wistar rats and 3 GK rats was extracted for whole genome resequencing on the BGI Illumina HiSeq 2500 platform. Then, align the truncated and filtered clean reads to the BN rat reference genome (w...

Abstract

The invention discloses SNP markers for identification of rats from four commonly-used lines and application thereof. The SNP markers includes 13 SNP sites, which are SNP1 to SNP13 that respectively indicate that the 13 SNP sites are located at position 201 of nucleotide sequences shown in SEQ ID No. 1 to SEQ ID No. 13. According to the invention, the advantages of high-throughput sequencing and biological information analysis technologies are adopted to develop the SNP markers which can be used for rapid identification of rats from four commonly-used lines (i.e., Wistar, GK, BN and SD). The application of the SNP markers can greatly reduce the genetic detection cost of rats and simplify operation.

Description

technical field [0001] The present invention relates to the use of high-throughput sequencing technology, bioinformatics analysis technology, generation sequencing technology and DNA mixing pool method to screen and verify 13 SNP markers of rats to quickly identify four commonly used strains (Wistar, GK, BN, SD) rats. Background technique [0002] In biomedical research, the genetic quality of experimental animals is an important factor to measure its quality. The use of experimental animals with stable genetic characteristics can ensure that the experimental results have more reference value and comparability. Therefore, in the process of raising experimental animals, regular genetic testing is an essential link. Traditional genetic quality detection methods such as biochemical marker gene detection, immune marker gene detection, and coat color gene detection have limitations such as low accuracy and few detection sites. These detection methods no longer meet the needs of ...
