Problems solved by technology

In particular, sample regions of interest may be missed by the initial low-resolution imaging scan, which are then not analyzed in subsequent scans and thus will not be analyzed at all
[0008] Furthermore, the use of matrix-assisted laser desorption ionization ("MALDI") techniques requires time-consuming sample preparation steps for matrix deposition
This step can introduce variability in the experiment because the matrix can vary within and between samples and can be unstable on the time scale of the experiment

Embodiment Construction

[0212]Embodiments disclosed herein provide methods and apparatus for imaging a sample in which the sample is measured in a first mode of operation by directing a spray of charged droplets onto the sample (i.e., performing a "full spectrum scan" to produce a "measurement image"), one or more regions of interest in the sample are identified based on data obtained from a full-spectrum scan (or based on analysis of a measured image), and then performed by directing a spray of charged droplets onto the sample (ie, subsequent " "analyzing a scan" to generate an "analyzing image") to analyze one or more regions of interest in a second, different mode of operation.

[0213] According to various embodiments, the overall analysis time of a sample may be reduced by identifying one or more regions of interest in an initial full-spectrum scan and then analyzing only the identified regions of interest in an analysis scan. For example, an initial full-spectrum scan may generate data represen...

Abstract

A method of analysing a sample is disclosed that comprises surveying a sample in a first mode of operation by directing a spray of charged droplets onto the sample, determining one or more regions ofinterest in the sample, and analysing the one or more regions of interest in a second different mode of operation by directing a spray of charged droplets onto the sample. The spot size of the spray of charged droplets at a point of impact with the sample may be varied.

Description

[0001] Cross References to Related Applications [0002] This application claims priority to and benefit from UK Patent Application No. 1609747.9 filed 3 June 2016, the entire content of which application is incorporated herein by reference. technical field [0003] The present invention relates generally to mass spectrometers, and in particular to methods of imaging samples using mass spectrometers. Background technique [0004] In mass spectrometry imaging, the spatial distribution of a sample's constituents is visualized by analyzing ions generated from multiple spatially separated regions of the sample. [0005] Mass spectral imaging of samples can be time consuming. For example, analysis of samples deposited on typical glass slides takes hours or even days. [0006] US Patent No. 7,655,476 (Bui) discloses a method for shortening the analysis time of mass spectrometry imaging. An initial tissue imaging scan is performed to obtain a relatively low-resolution mass spect...
