Problems solved by technology

However, like most separation methods based on cell density or size, surface acoustic wave cell separation technology has high efficiency in the case of large differences in cell size, but it is greatly limited in the separation of specific white blood cell subtypes. In particular, mononuclear cells (including lymphocytes and monocytes) in peripheral blood have similar densities (1.05–1.07 g cm −3 ), and the cell size is also relatively close to

Embodiment Construction

[0033]In order to make the object, technical solution and advantages of the present invention clearer, the present invention will be further described in detail below in conjunction with the accompanying drawings and embodiments. It should be understood that the specific embodiments described here are only used to explain the present invention, not to limit the present invention.

[0034] Aiming at the clinical requirement for convenient and accurate sorting and counting of white blood cell subtypes, the present invention proposes a microfluidic method combined with acousto-optic to realize label-free accurate white blood cell subtype sorting in the chip. The present invention is mainly made up of three modules, as figure 1 As shown, the three main modules are the surface acoustic wave focusing module, the surface acoustic wave sorting module, and the optical force sorting module. When sorting granulocytes, the surface acoustic wave sorting module is turned on, while the opti...

Abstract

The invention discloses an acousto-optic microfluidic chip for accurately sorting a leukocyte subtype and a sorting method therefor. The chip comprises a microfluidic channel, a surface acoustic wavefocusing module, a surface acoustic wave sorting module, and a light power sorting module. The surface acoustic wave focusing module processes the focused blood sample. The on-state of the surface acoustic wave sorting module and the light power sorting module is selected according to the type of a target cell to be sorted. The sorted blood sample flows into the corresponding collection bottle through a sample outlet. The surface acoustic wave cell is sorted and integrated into a microfluidic chip together with a laser photocell after performing the three-dimensional focus and separation, thereby greatly reducing the sizes of the instruments and overcoming the shortcomings of traditional instruments. The invention combines the advantages of the two techniques to accurately separate three leukocyte subtypes from peripheral blood without marking damages. The obtained three leukocyte subtypes have higher purity than 95%. The whole instrument is small, high precision and easy to operate, and has great application prospects.

Description

technical field [0001] The invention relates to the technical field of leukocyte subtype sorting, in particular to an acousto-optic microfluidic chip for accurately sorting leukocyte subtypes and a sorting method thereof. Background technique [0002] In clinical diagnosis, accurate sorting of immune leukocytes plays a key role in disease diagnosis and analysis, especially the precise separation of specific subtypes of white blood cells is an important prerequisite for in-depth understanding of disease occurrence and realization of disease treatment. For example, T cells, which play an important role in tumor immunity, require precise purification and sorting of T cells for functional analysis and research in the immune process, as well as promising tumor immunotherapy in tumor treatment in the future. for further analysis or cell proliferation. Traditional clinical methods are mostly based on centrifugation or electrochemical methods. Sorting cells according to their densi...
