Embodiment 1

[0029] The specific steps of the induction of Enterobacter sakazakii under the VBNC state of the present invention are as follows:

[0030] 1. Enrichment culture of Enterobacter sakazakii

[0031] Under sterile conditions, use an inoculation loop to pick out a single colony on the plate, inoculate it in LB broth medium and place it at 37°C for constant temperature cultivation for 18-24h. Pick a single colony on the plate and inoculate it in LB broth, and incubate at 37°C for 12h.

[0032] 2. Induction of Enterobacter sakazakii in VBNC state

[0033] (1) Induction of ampicillin

[0034] Take 1 mL of the bacterial solution cultured in step 1 and put it in a centrifuge for 2 minutes, discard the supernatant, add physiological saline to the precipitate and vortex. Dilute the respun bacterial solution 100 times, choose 10 7 CFU / mL concentration bacteria were induced, and the induction conditions were: temperature 24°C, humidity 80%. Add ampicillin to different centrifuge tube...

Abstract

The invention provides a rapid-induction forming method for VBNC-state enterobacter sakazakii. The rapid-induction forming method includes the steps that the enterobacter sakazakii is inoculated to becultured to the logarithmic phase, and a culture solution is obtained; then antibiotics and the culture solution are induced and cultured, wherein in induction and culture operation, the culture solution is taken and centrifuged, sediments are taken, resuspension is conducted, and the culture concentration is induced to 108 CFU / mL or below; the antibiotics are ampicillin, and the concentration ofthe antibiotics in the culture solution after induction culturing is 50 micrograms / mL to 1,600 micrograms / mL. By means of the rapid-induction forming method, the enterobacter sakazakii can be rapidlyinduced to be in a VBNC state, it is verified that the VBNC-state enterobacter sakazakii can be recovered with the conventional culture recovery method, and the pathogenicity can be recovered. Therefore, the rapid-induction forming method can lay the foundation for increasing the detection accuracy of the enterobacter sakazakii in milk products, and potential risk caused by existing of the VBNC-state enterobacter sakazakii is effectively avoided.

Description

technical field [0001] The invention belongs to the technical field of microorganism culture and detection, and more specifically relates to a method for rapid induction and formation of Enterobacter sakazakii in a VBNC state. Background technique [0002] The "viable but non-culturable state" (VBNC) of bacteria is a self-protection state that bacteria enter by adjusting their own metabolic pathways when they are stressed by an adverse external environment. During this process, the morphology of the bacteria gradually changes and cannot grow and reproduce on the plate. Therefore, the bacteria in the VBNC state cannot be detected by the conventional plate detection method. However, when the adverse external environment is removed, the bacteria in the VBNC state will recover and regain their cultivable ability. For some pathogenic bacteria, the pathogenic bacteria in the VBNC state are usually not pathogenic, but the pathogenicity of the pathogenic bacteria can be restored wi...
