Abstract

The invention discloses a preparation method of a thermally responsive bladder extracellular matrix bioactive hydrogel, relates to the field related to bioactive hydrogel preparation methods, and solves the problems that most of existing methods for obtaining an extracellular matrix (ECM) in the prior art have the defects of insufficient elution and large damage to ECM, and preparation of bladder-derived ECM hydrogels has not been reported so far. The preparation method comprises the following steps: step 1: taking a fresh pig bladder, keeping the ureter and urethral structure, and conductingrinsing with 3000 mL of double distilled water for 2 h; step 2: after wrapping by tin foil paper, quickly place the wrapped object in a refrigerator of -80 DEG C for 1 hour, then taking the object outand balancing the object to room temperature; step 3: repeating the step two for two times; step 4: conducting perfusion and immersing by double distilled water for 4 h, and immersing the whole bladder in a perfusate; and step 5: conducting perfusion and immersing by 1% Triton X-100 for 4 h, and immersing the whole bladder in a 1% Triton X-100 perfusate, and then conducting rinsing by double distilled water for 20 min.
