The annulus protection fluid comprises, by mass percentage, 2-10% of organic phosphine, 1-8% of a polymer dispersant, 5-15% of a corrosion inhibitor, 2-10% of a sterilizing agent, 1-5% of a solubilizing agent, 10-20% of a buffering agent, 2-10% of a deoxidant and the balance water. The method includes the steps that (1) the raw materials are weighed separately according to the ratio, the solubilizing agent and the buffering agent are mixed and stirred to be uniform, and then the mixture is added todeionized water with the temperature of 35-40 DEG C to obtain a mixture I; (2) the deoxidant, the sterilizing agent, the polymer dispersant, the corrosion inhibitor and the organic phosphine are sequentially added to the mixture I obtained in step (1), and stirring is performed for 1.5-2 hours at a constant temperature to obtain the oil field annulus protection fluid.
