Problems solved by technology

Although existing technologies such as in situ hybridization can study the gene expression of tissue cells in space, they still have many shortcomings, for example, they can only study the expression of some known genes, the procedures are complicated, and the resolution is too low, etc.

Abstract

The invention discloses a space transcriptome sequencing method. According to the method, space transcriptome information being high in resolution from tissue slices can be obtained, in situ sequencing is performed on a semiconductor sequencing chip for a space coding sequence connected to microspheres, and firstly, the corresponding relation of the space coding sequence on the microspheres and the micropore position of the sequencing chip can be confirmed, and subsequently, the tissue slices are attached to the chip loaded with the coding microspheres. Messenger ribonucleic acid mRNA in tissue can be caught in site, through reverse transcription, complementary deoxyribonucleic acid cDNA can be synthesized, and in the process of library construction and sequencing analysis, the sequence information of DNA can be obtained. Through software analysis, the sequencing results can be positioned to original positions in tissue, so that the method is favorable for gene expression of cells in research tissue and tissue pathology molecular mechanisms, and establishes an important foundation for comprehensive understanding and research of functions of tissue organs and disease evolution.

Description

technical field [0001] The invention relates to a spatial transcriptome sequencing method, in particular to a tissue section spatial transcriptome sequencing method based on a semiconductor chip for detecting pH value changes. Background technique [0002] The transcriptome information of cells in tissues is an important basis for studying the gene expression of cells, which can be used to study cell phenotype and function, reveal biological processes in tissues and molecular mechanisms in the process of disease occurrence. The various functions of complex tissues are closely related to the cell types in them, so the analysis of tissue cell transcriptomics is an important means to study the functions of tissues and organs and the vital activities of living organisms. [0003] At this stage, the transcriptome analysis of cells in tissues is usually a homogenized RNA sequencing analysis after biopsy tissue sampling. This method will result in the final transcriptome informatio...
