Problems solved by technology

In terms of bioinformatics, some scholars have studied the mechanism of PPARγ in many animal models and related diseases based on RNA-Seq, but the effect of exogenous expression of PPARγ on liver steatosis and lipid metabolism gene expression in mice has not been studied see the report
At present, thiazolidinediones (PPARγ agonists) have been widely used in the clinical treatment of type 2 diabetes, metabolic syndrome, immune diseases, etc., but due to the water and sodium retention, bone loss and heart failure risk caused by them , limiting the general clinical application of PPARγ receptor agonists

[0033] In this study, SPF grade C57BL / 6J mice aged 5-6 weeks and weighing about 20 g were used as animal models. The experimental mice were kept in the SPF grade animal room of the Medical Experimental Animal Center of the Medical Center of Xi'an Jiaotong University. The mice were given 12h light / dark alternation and had free access to drinking water.

Abstract

The invention discloses a research method for influence of a heterogenously expressed PPARgamma (peroxisome proliferator-activated receptor gamma) upon liver fatty degeneration and lipid metabolism related genes. The method comprises the following steps: dividing experiment mice into three groups, respectively performing tail intravenous injection of a LacZ adenovirus 4d, a PPARgamma adenovirus 2dand a PPARgamma adenovirus 4d, and killing the mice to obtain liver tissue; making the liver tissue into paraffin sections, performing H&E dyeing, making frozen sections, performing oil red O dyeing, extracting total RNA (ribonucleic acid) of the liver tissue, and performing gene chip analysis; and performing H&E dyeing and oil red O dyeing, and observing liver tissue lesion degrees; and performing gene chip analysis, so as to screen genes with differential expression caused by the heterogenously expressed PPARgamma in livers. By adopting the method, certain theoretic bases are provided for functions of the PPARgamma in livers and treatment on liver diseases.

Description

technical field [0001] The invention relates to the field of lipid metabolism, in particular to a research method for the influence of exogenous expression of PPARγ on hepatic steatosis and lipid metabolism-related genes. Background technique [0002] In recent years, the role of PPARγ in the liver has aroused people's high attention and extensive research. In terms of bioinformatics, some scholars have studied the mechanism of PPARγ in many animal models and related diseases based on RNA-Seq, but the effect of exogenous expression of PPARγ on liver steatosis and lipid metabolism gene expression in mice has not been studied See report. At present, thiazolidinediones (PPARγ agonists) have been widely used in the clinical treatment of type 2 diabetes, metabolic syndrome, immune diseases, etc., but due to the water and sodium retention, bone loss and heart failure risk caused by them , limiting the general clinical application of PPARγ receptor agonists. With the development...
