Abstract

The invention discloses a novel animal perfusion method. The method comprises the following steps: preheating a perfusion system; preparing the perfusion system by starting circulating water baths ofall components provided with a water jacket before an operation, ensuring that pipelines are clean, and replacing the pipelines when necessary; limiting the speed of perfusion liquid; performing vascular catheter insertion operation; and arranging the perfusion system. A model provided by the invention is a very useful tool for researching pathological processes and drug delivery mechanisms, migration/metastasis of circulating tumor cells into tumors, and interaction between an immune system and perfusion organs and tissues; interference caused by metabolic tissues such as liver, kidney, spleen and the like on experiments can be effectively avoided; and moreover, the invention provides a brand-new research approach with smaller interference for drug research and development, so that drug and tumor researchers can more intuitively know the action modes and effects of drugs on tumor tissues.

Meanwhile, long-term perfusion settings for bone, bladder, skin, prostate, and reproductive organs have not been described. Here, we report for the first time an in situ lower body perfusion protocol that bypasses the major clearing organs (kidney, liver, and spleen).
