Abstract

The present invention provides a kind of technique of the integration of rooting and transplanting of Mandshurica mandshurica tissue culture seedlings. First, the semi-lignified adventitious shoots obtained by tissue culture are placed in strong seedling medium (WPM+2.0mg / L 6-BA+ 20g / L sucrose+7g / L agar) after culturing for 30d, directly planted into the matrix (peat soil: vermiculite) containing rooting medium (WPM+1.4mg / L IBA+0.7mg / L NAA, without sucrose and agar) =3:1) in a seedling-growing bag, then sealed with a plastic pull bag, cultivated under specific light, temperature and humidity conditions, opened the bag mouth after 7 days, removed the bag after 14 days, and after 30 days of constant temperature and humidity cultivation, adventitious branches took root and grew out. The survival rate of new leaves is 63.33%. The invention simplifies rooting and transplanting into one step, which is one of the key technologies for rapid propagation of Mandshurica mandshurica seedlings, saves time and cost, and provides a large-scale method for the rapid propagation of Mandshurica mandshurica.
