Abstract

The invention discloses a plasma exosome miRNA biomarker and application thereof, wherein the miRNA biomarker comprises one or more of miR-15a-5p, miR-106b-5p and miR-107, and has the application in preparation of a kit for screening and diagnosing endometrial cancer. In 202 test samples, endometrial cancer patients and healthy volunteers (AUC = 0.873) can be distinguished through the expression quantity of three exosome miRNAs, and the exosome miRNAs can be combined with tumor markers CEA and CA125 (AUC = 0.904) to achieve higher sensitivity and specificity. Meanwhile, the three miRNAs also play a role in early diagnosis (diagnosis of a patient with endometrial cancer in the stage I), the joint AUC value of the three miRNAs in an ROC curve is 0.869, and the joint AUC value can be furtherimproved to 0.915 after the three miRNAs are combined with a tumor marker. Therefore, the kit prepared by the invention is composed of specific amplification primers and a general PCR amplification reagent, has high clinical value, and can be effectively used for screening and diagnosing endometrial cancer, including early screening.
