[0022] 2.1 Circ-RNF13 can combine with TRIM41 protein and p53 protein to form a tripartite complex

[0023] In order to further analyze the molecular mechanism of circ-RNF13, we confirmed that both TRIM41 and p53 can bind to circ-RNF13 through RNA pull down experiments combined with silver staining ( figure 1 a). The results of RIP experiments showed that in the group overexpressing circ-RNF13, TRIM41 protein and p53 protein combined more circ-RNF13 ( figure 1 b). RNA pull...

Abstract

The invention discloses the application of the circ-RNF13-TRIM41-p53 complex as a target site detection agent in the preparation of paclitaxel sensitization drugs for nasopharyngeal carcinoma. The study of the present invention shows that circ‑RNF13 in nasopharyngeal carcinoma paclitaxel-resistant cell lines can combine with ubiquitin ligase TRIM41 and p53 protein to form a complex to regulate nasopharyngeal carcinoma paclitaxel sensitivity. Mechanistic studies have shown that p53 protein mainly binds to the 1-176 nt region of circ-RNF13, and TRIM41 protein mainly binds to the 301-425 nt region of circ-RNF13. Blocking the related region of circ-RNF13 can make p53 protein bind to TRIM41 The binding ability of the protein is weakened, which weakens the ubiquitination and degradation of p53 by TRIM41, which in turn leads to the increased sensitivity of nasopharyngeal carcinoma cells to paclitaxel. Therefore, the circ‑RNF13‑TRIM41‑p53 complex can be used as a target site in the preparation of paclitaxel sensitizing drugs for nasopharyngeal carcinoma.

Description

technical field [0001] The invention belongs to the technical field of biomedicine, and specifically relates to the application of a circ-RNF13-TRIM41-p53 complex as a target site detection preparation in the preparation of paclitaxel sensitization drugs for nasopharyngeal carcinoma. Background technique [0002] Nasopharyngeal cancer (NPC) is a highly aggressive squamous cell carcinoma that is common in East Asia (especially southern my country), Southeast Asia and North Africa. In addition to genetic and environmental factors, Epstein-Barr virus (EBV) also plays an important role in the occurrence and development of nasopharyngeal carcinoma. Due to the complex anatomical structure of the nasopharynx and the high sensitivity of nasopharyngeal cancer cells to radiation, radiotherapy is the main treatment for nasopharyngeal carcinoma. However, due to the insidious onset of nasopharyngeal carcinoma, about 60% to 70% of patients are already in stage III or IV when they are dia...
