Embodiment 1

[0020] 1. Selection of explants and induction of clustered buds Wash the top centers of local red-skinned and white-skinned Jerusalem artichokes with tap water for 1-2 hours respectively, then rinse with sterile water once, and soak in 75% alcohol for 30 seconds , rinsed 3 times with sterile water, then soaked with 0.5% sodium hypochlorite for 5 minutes, rinsed with sterile water 3 times, each time for 5 minutes, and finally inoculated 1-5mm stem tips in 1- No. 20 culture medium, and then put on 3000lx light, 16h, 25°C, 8h dark culture, and culture at 20°C.

[0021]

[0022] In about 10 days, the white-skinned and red-skinned varieties all induced clustered buds on No. 7, No. 9, No. 13, and No. 15 media.

[0023] 2. Rooting culture Transfer the induced clustered shoots to MS medium or MS+NAA0.5 medium, culture at 3000lx light, 16h, 25°C, 8h dark, 20°C. Roots were induced after one week.

[0024] 3 Jerusalem artichoke shoot tip induced seedlings in one step. The white-skin...

Abstract

The invention provides a technique for the tissue culture, rapid propagation, domestication and transplantation of Helianthus tuberosus. The technique comprises the following steps of taking Helianthus tuberosus stem apexes as explants to induce cluster buds, inducing the cluster buds to root, inducing, transplanting and domesticating tuber buds and finally transplanting the tuber buds into field, so as to obtain Helianthus tuberosus seedlings. The method of the invention has the advantages that the method is simple, easy to master, applicable to different varieties of Helianthus tuberosus, high in the value-added coefficient of the cluster buds and the tuber buds of tissue culture seedlings, orderly in rooting, sturdy in seedlings, high in survival rate and applicable to the commercial production of Helianthus tuberosus seedlings.

Description

Technical field: [0001] The invention belongs to the field of biotechnology. In particular, it relates to tissue culture rapid propagation and domestication transplanting techniques of Jerusalem artichoke. Specifically, it involves a complete production technology of taking Jerusalem artichoke stem tips as explants, obtaining secondary buds through organogenesis, and multiplying, rooting, domesticating and transplanting in large quantities: a method for tissue culture, rapid propagation and transplanting of Jerusalem artichoke. Background technique: [0002] Jerusalem artichoke (Helianthus tuberosus L.) belongs to the perennial herbaceous plant of the genus Sunflower in the family Asteraceae, also known as Jerusalem artichoke and ghost ginger. Jerusalem artichoke is native to Brazil and was introduced to France, Italy, Japan and China after the 17th century. Jerusalem artichoke has a wide distribution range in my country and can grow from south to north. Jerusalem articho...
