Problems solved by technology

But on pigs, due to the difference between its hair characteristics and human beings, the hair is thick and hard, with grease and other sundries, making the processing process more complicated, and because of the differences between species, the cortisol content in pig hair is not the same as that of humans. Similarly, there is no report on the determination of cortisol content in pig hair

Embodiment 1

[0025] 1) Select 5 Taihu pigs and Landrace pigs with similar weight and age, and collect pig hairs from the same part of the neck respectively. The length of the pig hairs is 0.8-1.2 mm; each takes 200 mg of pig hairs and washes them with 10 ml of anhydrous methanol 3 times, each time no more than 3 minutes, can be shaken and washed on the shaker;

[0026] 2) Dry the washed pig hair and mix it with 1ml of anhydrous methanol to obtain a mixture;

[0027] 3) Rotate the mixture at 360° for 24 hours on a rotator with a speed of 30 rpm, and then put it into a centrifuge at a temperature of 20°C at a speed of 4500 rpm / min, and collect the supernatant after centrifugation for 15 minutes ;

[0028] 4) After the collected supernatant was concentrated and blown dry with nitrogen flow, it was dissolved in PBS solution (PH=7.4, 0.1M) with a volume of 150ul, and the cortisol content in the solution was measured by radioimmunoassay.

Abstract

The invention discloses a method for extracting and detecting cortisol from pig hair, belonging o the field of analysis and detection. The method comprises the steps: preparing pig hair, washing the pig hair by using an organic solvent, air-drying, putting the air-dried pig hair into a centrifuge tube filled with the organic solvent, and rotating the centrifuge tube on a rotator; putting the centrifuge tube into a centrifugal machine for centrifugation after the treatment, and collecting a supernatant liquid; and concentrating and blow-drying the supernatant liquid by using a nitrogen flow, dissolving the blow-dried supernatant liquid in a PBS (Phosphate Buffered Solution), and then measuring the content of the cortisol in the solution by adopting a radioimmunoassay method. The method is a way for detecting the physiological status of a pig without causing abuse and stress to the pig, is simple and easy to implement and low in cost, and plays a quite positive effect for monitoring the health and welfare conditions of the pig.

Description

technical field [0001] The invention belongs to the field of analysis and detection, and in particular relates to a method for extracting and detecting cortisol from pig hair. Background technique [0002] my country is a big pig raising country, but the long-term blind pursuit of "fast" and "high yield" in the breeding industry has brought about a series of serious environmental and social problems, such as the deterioration of the breeding environment, serious environmental pollution, frequent outbreaks of livestock and poultry epidemics, This not only causes huge economic losses, but also endangers human health. Under irregular breeding or high-intensity intensive farming, pigs are often subjected to various stresses (weaning, mixing groups, castration, transportation, etc.), and long-term stress will eventually reduce the immunity and resistance of pigs. Often rely on the addition of excessive antibiotics and various vaccines to maintain animal health or life, which not ...
