[0024] Example: Regeneration of functional human skin and hair follicles in nude mice using in vitro culture-expanded cells

[0025] Materials and methods:

[0026]Animals: Immunodeficient nude mice (or homozygous nu / nu mutant mice) or non-diabetic obese mice / severe combined immunodeficiency mice are albino mice. Therefore, it is suitable for various xenograft tissue transplantation and tumor transplantation.

[0027] Cells and Cell Culture Media: Dissociated epidermal cells were obtained from human neonatal foreskins. Dermal cells come from the scalp of a 15-week-old fetus. After the cells were isolated from the tissue, they were cultured in dermal cell medium DMEM / F12 (1:1) (Gibco, Cat. #11039). Epidermal cells were cultured in CNT07 medium containing protein kinase (ROCK) inhibitor Y27632. The dermal cells and epidermal cells need to be cultured to the third cycle and the third generation respectively. Then mix at a ratio of 1:1 to make 150 μl of cell suspension DMEM / F...

Abstract

The invention discloses a skin model and application. The method comprises separately culturing epidermal cells and dermal cells derived from fetus scalp, newborn prepuce or adult scalp, and subculturing, wherein the used culture medium and culture method can maintain multiple functions of skin cells; separately digesting the cultured and proliferated multifunctional epidermal cells and dermal cells into single cells, and mixing to obtain suspension; transferring onto a semipermeable polymer or silica gel membrane substrate for incubation; transplanting cells on adhesive film to a receptor. The method can regenerate human intact functional skin and hair, the regenerated skin contains an epidermal layer, a dermal layer and a subcutaneous tissue as well as skin appendant organs. The skin model regenerated by the method disclosed by the invention can be used for study of human skin diseases, screening of skin drugs, and development of cell products for treating wound and other skin diseases.

Description

technical field [0001] The present invention relates to a method for regenerating intact human skin tissue with cells cultured in vitro, the present invention also relates to the skin and hair model prepared by the method, and the use of the model in the research of human skin and hair diseases, the screening and development of skin medicines Applications in clinical products for the treatment of trauma and baldness. Belongs to the field of histocytology and tissue engineering. Background technique [0002] Dermatologists and skin surgeons have long struggled to regenerate fully functioning skin clinically for the healing of skin trauma and the removal of skin scarring. There are many reasons for skin damage: such as genetic diseases, burns, chronic trauma such as trauma caused by diabetes. Many skin damages cannot be recovered by themselves, such as deep large-scale burns, and large-area damage to the skin can easily lead to infection and death. At present, the most comm...
