Problems solved by technology

This method simplifies the process of processing plant materials, resulting in large errors in the determination of lignin content.
In addition, except for the above-mentioned several invention patents related to the method of measuring lignin in plants, there are few or no patents on other similar methods of measuring lignin, which shows that there are still many problems in the method of lignin determination that need to be further studied

Embodiment Construction

[0032] The method for measuring the lignin content of herbaceous plants comprises the following steps:

[0033] (1) Dry the collected plant samples at 80°C for 48-72 hours, use a miniature plant grinder to crush the plant samples, pass through a 30-mesh sieve after crushing, and collect the plant sample powder.

[0034] (2) Weigh 0.05~0.10g of plant sample powder, and record the specific value as n; then put the plant sample powder into a 50mL centrifuge tube, add 10mL of acetic acid solution with a mass concentration of 0.1%, stir it evenly with a glass rod, and place it for 20 After ~30min, fully softened plant sample powder is obtained.

[0035] (3) Cover the centrifuge tube with the fully softened plant sample powder inside, put it into the centrifuge, and centrifuge at 3500rpm for 5min to obtain the supernatant A and sediment A respectively; pour off the supernatant A, and use the mass concentration Wash precipitate A with 5 mL of 0.1% acetic acid solution.

Abstract

The invention relates to a method for measuring the lignin content of herbaceous plants. The method comprises the following steps: (1) drying, smashing and sieving a plant sample to obtain plant sample powder; (2) adding acetic acid to the plant sample powder to obtained fully-softened plant sample powder; (3) carrying out centrifugation to obtain a precipitate A; using the acetic acid to clean the precipitate A; (4) embathing the precipitate A into an ethanol-ether solution to obtain a precipitate B; (5) after the precipitate B is dried by distillation, adding sulfuric acid for stirring to be uniform, and then standing at room temperature; (6) adding distilled water for stirring to be uniform; after boiling water bath and cooling at the room temperature, sequentially adding the distilled water and barium chloride to shake up for centrifugation to obtain a precipitate C; (7) after the precipitate C is flushed with water, sequentially adding the sulfuric acid and potassium dichromate, and cooling to the room temperature after reaction; (8) transferring a substance obtained in the step (7) into a conical flask; (9) after adding potassium iodide into the conical flask for reaction, adding a starch indicator, using a sodium thiosulfate solution to titrate to a terminal point; (10) carrying out a blank test; (11) calculating the lignin content. The method is simple, convenient and accurate.

Description

technical field [0001] The invention relates to a measuring method, in particular to a measuring method for the lignin content of herbal plants. Background technique [0002] As we all know, lignin is the second organic polymer compound after cellulose in nature. Lignin is mainly distributed in the tubular molecules and fibers of the xylem in plants, sclerenchyma cells, sclerenchyma cells, and the secondary walls of specific types of epidermal cells, and constitutes the three main components of plant skeletons together with cellulose and hemicellulose. The body mainly plays the functions of mechanical support, preventing biodegradation, disease defense, and transporting water. Lignin is formed by the polymerization of three types of phenylpropenyl alcohols (single woody alcohols): coumarly alcohol, coniferly alcohol and sinapya alcohol. Because lignin monomers are different, lignin can be divided into three types: syringyl lignin (S-lignin) polymerized from syringyl propan...
