Embodiment
[0015] Weigh 8.4g of lithium hydroxide monohydrate and 8g of urea, dissolve them in 83.6g of water, and filter to obtain an aqueous solvent. Add 1 g of chitosan with a degree of deacetylation of 85% and a molecular weight of 600,000, stirring and dissolving at low temperature. Add 10 mL of butylene oxide to the chitosan solution and stir at 25°C for 10 h. The reaction solution was neutralized to neutral with hydrochloric acid, loaded into a dialysis bag for dialysis for three days, and then freeze-dried to obtain hydroxybutyl chitosan. The phase transition temperature of the 3wt% aqueous solution of the product is 51 degrees Celsius.

The invention relates to a new method for preparing temperature-sensitive hydroxybutyl chitosan. A chitosan water solution is obtained by directly dissolving chitosan in an aqueous solvent at low temperature. The chitosan water solution further reacts with epoxybutane at room temperature. Inorganic components and unreacted impurities in a reaction solution are removed by dialysis, and white water-soluble hydroxybutyl chitosan can be obtained after freeze-drying. The derivative has the nature of being gelled when heated and being re-dissolved when cooled. Products with different phase transition temperatures can be obtained by controlling the feed ratio and reaction time. The hydroxybutyl chitosan prepared by utilizing the method has great water solubility. The preparation method is simple and easy to operate, the use of organic solvents can be avoided, and the reaction time can be greatly shortened simultaneously. As organic solvents are not used at all, the method has little influence on the environment. The hydroxybutyl chitosan prepared by the method provided by the invention can be used for the fields of medicament delivery, injection type tissue engineering scaffolds, bio-sensing and the like.
Description
technical field [0001] The invention relates to a preparation method of temperature-sensitive hydroxybutyl chitosan, which belongs to the field of natural biopolymer materials and also belongs to the field of biomedical materials. Background technique [0002] Chitosan is obtained by the deacetylation of natural polymer chitin. It has excellent biocompatibility, biodegradability and natural antibacterial properties. It is widely used in biomedical fields such as drug delivery, tissue engineering, and wound accessories. Applications. [0003] As a semi-crystalline polymer, chitosan has a relatively regular chain structure. Therefore, it is difficult to dissolve in water and common organic solvents, so the application of chitosan is generally dissolved in a slightly acidic environment. However, some applications are not suitable for acidic environments, and some chitosan modification reactions are not suitable for acidic conditions. It is difficult for the acidic solution o...
