By simultaneously adding primers CMCT505 and CMTA170a and adjusting concentrations of DNTPs, Buffer and a template DNA, an annealing temperature and the like, orthogonal tests of various affecting factors are designed, by identifying DNA of polymeric gene seedlings obtained by amplifying and screening bi-parental progeny F1 and a single PCR system, the dual-SSR-PCR system capable of simultaneously amplifying 189bp and 121bp two target strips is established.
