Embodiment 1

[0022] 1. Material: Coniogramme japonica (Thunberg) Diels.

[0023] 2. Material category: fern spores that grow naturally in the wild.

[0024]3. Culture conditions: (1) Spore germination medium: the basic medium is 1 / 4 of MS macronutrients, and 1 / 4MS medium with the same ingredients as MS; (2) GGB induction and proliferation medium: MS+6 -BA0.5mg / L+NAA0.2mg / L; (3) rooting medium: 1 / 10MS; (4) differentiation medium: 1 / 2MS+IBA0.1mg / L+IAA0.1mg / L; (5) In vitro storage medium: 1 / 2MS. All the above mediums were added with 30g / L sucrose, solidified with 5.8g / L agar, pH5.8, the culture temperature was 25±2℃, and the light intensity was 10-20μmol / (m 2 s), light 12h / d. The culture bottles used are white transparent glass bottles with a diameter of 7.5 cm x a height of 9.0 cm and a volume of about 300 ml. All media were solidified by adding agar 5.8g / L, pH5.8. (1)(2)(3)(5) Culture under the following culture conditions: culture temperature 23±2℃, light intensity 10-20μmol / (m 2 s),...

Abstract

The invention provides a rapid propagation and in-vitro preservation method of a plant coniogramme japonica (Thunberg) diels. The rapid propagation and in-vitro preservation method comprises the steps of obtaining an explant, carrying out green globular body GGB induction and propagation culture, carrying out rooting culture, carrying out differentiation culture and carrying out in-vitro preservation or transplanting, i.e., taking coniogramme japonica (Thunberg) diels spores which naturally grow in the wild; culturing in a spore germination culture medium to obtain seedlings with root stocks; transferring the seedlings into a green globular body induction and propagation culture medium for culturing, and transferring generated green globular bodies into a rooting culture medium; transferring the green globular bodies which have 1 to 2 roots, have differentiated leaflets and are not completely differentiated into a differentiation culture medium for culturing; transferring the differentiated leaflets onto an in-vitro culture medium for subculture and in-vitro preservation; or transplanting the differentiated leaflets. The method provided by the invention provides important technical supports to development, utilization and protection of coniogramme japonica (Thunberg) diels plant germplasm resources.

Description

Technical field: [0001] The invention relates to the field of plant biotechnology, in particular to a method for in vitro preservation of the plant Coniogramme japonica (Thunberg) Diels. Background technique: [0002] Coniogramme japonica (Thunberg) Diels is a perennial herbaceous plant of the genus Coniogramme Fee in the family Hemionitidaceae. It is widely distributed in southwestern my country and the middle and lower reaches of the Yangtze River. It likes to grow in humid forests and damp places in valleys. , 100-1300 meters above sea level. Its underground rhizomes are developed and often grow in a row. The rhizomes and the whole herb are used as medicine, which has the effects of relaxing muscles and activating collaterals, dispelling cold and dehumidification, and is mainly used to treat rheumatoid arthritis, leucorrhea, nephritis, and unknown sores. At the same time, the plants of Fengya fern are beautiful in shape, and they are also excellent ornamental ferns. At p...
