Problems solved by technology

[0006] At present, there is no relevant application for identifying the sex of sparrows by molecular means, that is, there is no patent for gender identification based on specific primers of the NIPBL gene (Nipped-B protein homologous gene), and there are no successful examples of sparrows reported in the literature.

Embodiment 1

[0028] 1) Extraction of total genome: Using phenol and chloroform extraction method, take about 50 mg of muscle tissue sample numbered K0672 (see Table 1 below), cut it and place it in a sterilized 2.0 mL EP tube;

[0029] 2) Add 1mL of DNA extraction solution and 4μL of 20mg / mL RNaseA (ribonuclease) into the EP tube, shake well and place in a 37℃ water bath for 1h;

[0030] 3) Take out the EP tube, add 5μL of 20mg / mL proteinase K, shake well and place it in a water bath at 50℃ for about 2-3 hours until the muscle is completely digested, shaking up and down every 10 minutes during this time;

[0031] 4) Take out the EP tube, after cooling to room temperature, add an equal volume of saturated phenol, gently shake up and down for about 7 minutes until the water phase and the phenol phase are mixed into an emulsion;

[0032] 5) Centrifuge at 10000rPm for 15min at 10℃, and take the upper aqueous phase to another 20mL EP tube;

[0033] 6) Repeat phenol extraction once or twice;

[0034] 7) Ad...

Embodiment 2

[0046] Operate according to the method of Example 1, the difference is that the number of the sample to be tested is K0673 (shown in Table 1 below), and the electrophoresis results are as follows figure 1 As shown (lane number is 3).

Embodiment 3

[0048] The operation was carried out according to the method of Example 1, except that the number of the sample to be tested was K0663 (shown in Table 1 below), and the electrophoresis result was the same as that of Example 1 (the lane number was 5).

Abstract

The invention discloses specific gene primers for identifying sparrow gender and the application thereof. The primers include KLNIPBL-i16F2 with serial number of SEQ ID NO.1 and KLNIPBL-i16R2 with serial number of SEQ ID NO.2. Amplification of genomic DNA extracted from a sparrow meat sample or the blood of an adult sparrow is conducted by means of the pair of primers, agarose gel electrophoresis is conducted on an amplification product, the result shows that amplification sequence is the target fragment of NIPBL gene, a female sparrow and a male sparrow are obviously different in electrophoretic band, and sparrow gender can be clearly identified according to the number of bands. The method and technique are high in practicality and repeatability, and can accurately identify sparrow gender.

Description

Technical field [0001] The invention belongs to the technical field of biological detection, and specifically relates to a primer for identifying a specific gene of sparrow sex and its application. Background technique [0002] With the rise of gender identification research, identification methods are changing with each passing day. At present, the use of molecular biology methods to conduct gender identification analysis has become one of the necessary methods. Although traditional morphological judgment methods are common, it is not unreliable when judging sparrow chicks that have just hatched with the same color regardless of male and female. However, it is difficult to determine the male and female of sparrows from traditional morphological characteristics to conduct gender identification by molecular means, such as the samples are sparrow larvae or defects, corrupted tissues, etc. So a method for species identification using DNA sequences has emerged, which provides a con...
