[0042] 1. Culture effect of green flagellar cell culture stage

[0043] Algae were cultured on BBM medium. Inoculation initial OD 680 is 0.5. The culture period is 9 days. The culture temperature is 25±1℃, and the culture light intensity is 25μmol·m -2 ·s -1 , Continuous lighting, the light source is LED plant cultivation lights, white light. BBM culture reagent medicine: NaNO 3 , MgSO 4 ·7H 2 O, NaCl, K 2 HPO 4 、KH 2 PO 4 , CaCl 2 , ZnSO 4 ·7H 2 O, MnCl 2 ·7H 2 O, MoO 3 、CuSO 4 ·5H 2 O. CoNO 3 ·6H 2 O, H 3 BO 3 , EDTA, KOH, FeSO 4 ·7H 2 O, BBM medium components are shown in Table 1.

[0045]

[0046] After 9 days of culture, the dry weight of cells increased with time, as figure 1 As shown, the highest reaches 1.15g / L. After 9 days of culture, flagellar cells accounted for 91% of the total number of cells, spherical cells accounted for 6%, and dead cells accounted for 3%. The percentages of various types of cells i...

Abstract

The invention discloses a method for producing astaxanthin by haematococcus pluvialis rich in the astaxanthin, and relates to production and application of micro-algae. The method includes the steps:culturing flagellate cells of the haematococcus pluvialis; culturing spherical cells of the haematococcus pluvialis; accumulating haematococcus pluvialis astaxanthin. Physiological and biochemical characteristics of cells in various forms of the haematococcus pluvialis are sufficiently used, culture is reasonably performed by stages by the aid of the physiological and biochemical characteristics of the haematococcus pluvialis cells in different forms, so that cell biomass is improved, cell survival rate is increased, and the content of astaxanthin is improved. The flagellate cells of the haematococcus pluvialis are transformed into larger spherical cells, so that algae cells in algae liquid can be harvested by a natural sedimentation method of the algae cells, and harvesting cost is reduced.

Description

technical field [0001] The invention relates to the production and application of microalgae, in particular to a method for producing astaxanthin by using Haematococcus pluvialis rich in astaxanthin. Background technique [0002] Astaxanthin is a red ketone carotenoid with super antioxidant capacity (Guerin et al., 2003; Goswami et al., 2010), widely used in aquatic feed, cosmetics, food and health products and other fields (Johnson and An, 1991; Nelis and De, 1991; Guerin et al., 2003; Goswami et al., 2010; Tominaga et al., 2012; Higuera, 2006). Haematococcus pluvialis is a green alga capable of synthesizing and accumulating high levels of natural astaxanthin, which can reach 4%-5% of dry weight (Margalith, 1999; Boussiba, 2000; Chen, 2015) . The market demand for astaxanthin is huge. In 2014, the global astaxanthin market was about 447 million US dollars. It is estimated that by 2020, the global astaxanthin market will reach a scale of 1.1 billion US dollars. , Aquacult...
