The invention discloses a protein separation and extraction device which comprises three parts, namely material tanks, liquid collection tanks and chromatographic columns, wherein the liquid outlet ofthe first material tank is connected with the second material tank through a first liquid guide pipe; a waste liquor opening of the first material tank is connected with the first liquid collection tank through a third liquid guide pipe; the second material tank is divided into two storage chambers through a dialysis membrane arranged therein; the upper storage chamber is connected with the thirdmaterial tank through a second liquid guide pipe, and the lower storage chamber is connected with the second material tank through a fourth liquid guide pipe; the third material tank is divided intotwo protein storage chambers through a molecular sieve arranged therein; the upper protein storage chamber is connected with the top of the second chromatographic column through a fifth liquid guide pipe; the bottom of the second chromatographic column is connected with the fourth liquid collection tank; a bottom opening of the lower protein storage chamber is inserted into the top of the first chromatographic column; the bottom of the first chromatographic column is connected with the third liquid collection tank. The protein separation and extraction device disclosed by the invention is highin efficiency, low in energy consumption and capable of avoiding addition of any chemical reagent on the basis of maintaining the protein activity.
