The preparation method ofthe PDMS template comprises the following steps: a, etching a cylindrical micro-column array, consisting of micro-columns, in an etching area on the surface of a silicon wafer to obtain a micro-columnsilicon wafer; b, carrying out hydrophobic treatment on the surface of the obtained micro-column silicon wafer; c, dropwise adding a PDMS material into the etching area where the micro-column array is located, and uniformly spreading; d, enabling the PDMS material to be solidified so as to form a PDMS thin film, and separating the PDMS thin film from the micro-column silicon wafer to obtain the PDMS template with micro-column through holes.
