The invention provides a method for assessing content of gas in shale based on methane carbon isotope, which estimates relevant parameters by matching of experimental results and calculations, and utilizes the change of methane carbon isotope value during desorption and deflation of shale gas to establish a corresponding relationship with the gas recovery rate during the deflation to achieve the calculation of a total gas volume. On the one hand, the method can calculate a lost gas volume, a desorbed gas volume and a residual gas volume, and on the other hand, can calculate a free gas volume and an adsorbed gas volume. On the one hand, the isotope fractionation provided by the invention can calculate a lost gas, a desorbed gas, and a residual gas; and on the other hand, can calculate a free gas and an adsorbed gas.
