Problems solved by technology

However, simply using the processing solution used in traditional serum testing is not enough to completely remove the interference factors in the sample, and the accuracy of the test results needs to be improved. Unnecessary clinical examination and unreasonable treatment in the later stage

Embodiment 1

[0047] A treatment solution for alveolar lavage fluid samples, comprising the following raw materials: treatment solution A (KCl 0.12M, Triton X-100 0.01%) and treatment solution B (KOH 0.25M).

[0048] The method for detecting fungi by pretreating the alveolar lavage fluid with the sample treatment solution, the specific steps are as follows:

[0049] Step 1: Prepare treatment solution A and treatment solution B respectively, and mix equal volumes of treatment solution A and treatment solution B to obtain the sample treatment solution;

[0050] Step 2: Add 40 μL of sample treatment solution to 10 μL of alveolar lavage fluid sample, mix well and incubate at 37°C for 10 min;

[0051] Step 3: Add 200 μL of the main reaction agent to the incubated mixture, shake and mix well, detect and calculate the content of (1-3)-β-D glucan.

Embodiment 2

[0053] A treatment solution for alveolar lavage fluid samples, comprising the following raw materials: treatment solution A (KCl 0.12M, Triton X-100 0.10%) and treatment solution B (KOH 0.25M).

[0054] The method for detecting fungi using the sample treatment solution to pretreat the alveolar lavage fluid is as in Example 1.

Embodiment 3

[0056] A treatment solution for alveolar lavage fluid samples, comprising the following raw materials: treatment solution A (KCl 0.12M, Triton X-100 0.50%) and treatment solution B (KOH 0.25M).

[0057] The method for detecting fungi using the sample treatment solution to pretreat the alveolar lavage fluid is as in Example 1.

Abstract

The invention discloses a bronchoalveolar lavage fluid sample treatment fluid and a treatment detection method. The treatment fluid consists of inorganic salt and a surfactant and can reduce interference of other substances in a bronchoalveolar lavage fluid to fungus (1-3)-beta-D-glucan detection. A bronchoalveolar lavage fluid sample is treated to be used for detecting the content of fungus (1-3)-beta-D-glucan in the bronchoalveolar lavage fluid of an invasive pulmonary fungal infection patient in vitro. Compared with a traditional serum detection method, the method is higher in detection sensitivity and can keep higher specificity and accuracy.

Description

technical field [0001] The invention belongs to the field of in vitro detection of fungi (1-3)-β-D-glucan, in particular to a treatment solution for alveolar lavage fluid samples and a treatment and detection method. Background technique [0002] The invention is used for auxiliary diagnosis of clinically invasive deep fungal infection. With the continuous development of clinical techniques such as corticosteroids, immunosuppressants, the extensive use of various broad-spectrum antibiotics, and organ transplantation, for some immunocompromised patients, the probability of invasive fungal infection will generally increase. (1-3)-β-D glucan is the cell wall component of most medically important fungi (including Candida and Aspergillus). The concentration in body fluids such as lavage fluid and cerebrospinal fluid increases, so the detection of (1-3)-β-D glucan is a key indicator. At present, most of the G test samples are serum. In contrast, in patients with invasive pulmona...
