[0048] The tissue samples used in this application study were all obtained from cardiac surgery patients in Qianfoshan Hospital Affiliated to Shandong University (Jinan, China). The specimens of the control group were taken from heart transplant donor volunteers (n=7), see Table 1A for details. Atherosclerotic tissue specimens were obtained from patients with aortic aneurysm and aortic dissection (n=7) with atherosclerotic conditions. Most of the patients had hypertension, and imaging tests showed aortic calcification, as shown in Table 1B. The samples were immediately stored in a -80°C refrigerator after being collected from relevant tissues for subsequent experiments. Abstract

The invention belongs to the field of anti-atherosclerosis drugs, in particular to the application of carbonic anhydrase inhibitors in the preparation of anti-atherosclerosis drugs. This study found for the first time that CA1 is highly expressed in atherosclerotic calcified tissue, and CA1 is associated with vascular smooth muscle calcification and affects cell proliferation and apoptosis. Methazolamide, an inhibitor of CA1, can significantly attenuate the process of atherosclerosis and inhibit the inflammatory factors of atherosclerosis. The above results suggest that calcification plays an important role in atherosclerosis, and CA1-dominated calcification promotes the process of atherosclerosis. Methazolamide has potential therapeutic effect on atherosclerosis by inhibiting CA1 expression. These findings not only further illustrate the close relationship between atherosclerosis and calcification, but also explore the calcification mechanism of the disease, and also prove that inhibiting calcification is the key point in the treatment of atherosclerosis. This study provides a new idea for the treatment of this disease.
