Problems solved by technology

During the freeze-drying process, when the temperature drops and the vacuum increases, the water in the cells freezes and evaporates, and the solutes and electrolytes in the cells are gradually concentrated, resulting in excessive water loss of cells, severe shrinkage and deformation, and even death; in addition, When the el...

Abstract

The invention provides a freeze drying method for improving the survival rate of lactobacillus plantarum and belongs to the field of freeze drying. The method provided by the invention comprises the following steps: step 1, performing strain activating to obtain a single colony; step 2, inoculating and culturing the single colony to obtain a seed liquid; step 3, transferring the seed liquid to anMRS liquid culture medium containing 0.5-2 g/L octadecenoic acid for expanding culture to obtain an expanded culture solution; step 4, centrifuging the expanded culture solution to obtain thallus precipitate; and step 5, washing the thallus precipitate with a PBS buffer solution, re-suspending the thallus precipitate in a PBS solution or a protective agent solution, transferring the thallus precipitate into a container, and freeze-drying the thallus precipitate. The octadecenoic acid is preferably oleic acid. The octadecenoic acid is used in the expanding culture step, so that the survival rate of the rehydrated lactobacillus plantarum after freeze drying is obviously improved, and unexpected technical effects are brought.

Description

technical field [0001] The invention relates to a freeze-drying method, in particular to a freeze-drying method for improving the survival rate of Lactobacillus plantarum, and belongs to the field of freeze-drying. Background technique [0002] Water is indispensable for the metabolism of the body and the maintenance of life processes. In the absence of water, living cells are dead or dormant. Therefore, in order to stabilize unstable products such as probiotics, the water content in stored samples must be reduced to a dormant state. Freezing is a unit operation that freezes moisture, thereby reducing the water content of the sample to a dormant state. However, maintaining and shipping samples in a frozen state is expensive and may result in loss of value of the product. Alternatively, samples can be dried in air at high processing temperatures, but traditional drying methods can cause changes in the physical and chemical properties of the sample. Freeze-drying just comb...
