Abstract

The invention discloses an aegilops biuncialis specific molecular marker obtaining method and an application thereof. The method comprises the following steps: extracting genome DNA of aegilops biuncialis to be detected, and then simplifying genome library construction and high-throughput sequencing; comparing an SLAF tag obtained by sequencing after sequence analysis and screening of the SLAF tagof a specific gene with a reference genome sequence of known wheat; and screening sequences of which homology is less than 50% for primer designing; screening specific molecular markers of aegilops biuncialis, further positioning aegilops biuncialis primers obtained by screening to specific genes in aegilops biuncialis by utilizing aegilops tauschii genome donors with specific genes in homologousaegilops tauschii, and performing PCR amplification on the designed primers to obtain the specific molecular markers of aegilops biuncialis. The method can be used for detecting genetic components such as aegilops biuncialis chromosomes and chromosome fragments under a wheat background, can be used for rapidly detecting the wheat-aegilops biuncialis derivative line material on a large scale by simplifying a genome sequencing technology, and is time-saving, labor-saving and low in cost.
