Puccinia malvacearum-resistant gene pmaeu1 in aegilops tauschii and molecular markers and application thereof

This invention relates to a powdery mildew resistance gene PmAeu1 derived from *Aegilops spp.*, its molecular markers, and applications, belonging to the field of genetic engineering. This invention utilizes the powdery mildew-resistant *Aegilops spp.* strain AE 1617 and the powdery mildew-susceptible strain AE 538 to construct mapping populations. Through BSR-Seq, fine genetic mapping, third-generation genome sequencing, and transgenic verification, a new powdery mildew resistance gene, PmAeu1, was cloned. This invention has demonstrated through transgenic wheat research that the PmAeu1 gene can significantly improve wheat resistance to powdery mildew. The *Aegilops spp.* powdery mildew resistance gene PmAeu1 can be applied to genetically improve wheat varieties to enhance wheat's powdery mildew resistance. This invention also develops a molecular marker, XAeuNLR1-2, for the PmAeu1 gene, which can be used to specifically detect the gene PmAeu1 as described in the claims and can be applied to marker-assisted breeding of the PmAeu1 gene.

Candidate gene AetNLR2CIae8 of aegilops tauschii powdery mildew resistance gene pmAeCIae82DS and application thereof

The invention discloses a candidate gene AetNLR2CIae8 of a powdery mildew resistant gene pmAeCIae82DS derived from aegilops tauschii as well as a protein coded by the candidate gene AetNLR2CIae8 and application of the candidate gene AetNLR2CIae8, and the nucleotide sequence of the candidate gene AetNLR2CIae8 of the powdery mildew resistant gene derived from the aegilops tauschii is as shown in SEQ ID NO. 1. 1, and amino acid sequences coded by the gene are respectively as shown in SEQ ID NO. 2. The pmAeCIae82DS gene sequence belongs to the protection range of the invention. The invention also protects the application of the candidate gene AetNLR2CIae8 of the pmAeCIae82DS in the improvement of the disease resistance of the plants. The AetNLR2CIae8 is transformed into the susceptible wheat variety Yangmai 158 by utilizing a transient overexpression system, so that the aspirator index of powdery mildew can be obviously reduced; when the VIGS system is used for respectively silencing AetNLR2CIae8 in the aegilops sphaeroides powdery mildew resistant material TX11-1 containing the pmAeCIae82DS, the susceptibility of plants can be improved. Therefore, the AetNLR2CIae8 can positively regulate and control the wheat powdery mildew resistance and is an important gene resource for wheat disease-resistant breeding.

Tissue culture method of aegilops tauschii

The invention discloses a tissue culture method of aegilops tauschii. Comprising the following steps: (1) disinfecting an explant, namely disinfecting a mature embryo of aegilops tauschii as the explant; (2) induction and subculture of calluses: inoculating the explants on an induction culture medium, and carrying out dark culture to induce embryogenic calluses; (3) differentiation of the calluses: transferring the calluses into a differential culture medium, and differentiating to obtain bud seedlings; (4) rooting: transferring the bud seedlings to a rooting culture medium to differentiate and root; and (5) hardening seedlings and transplanting after complete plants are formed. The efficient tissue culture regeneration system of aegilops tauschii is constructed for the first time, mature embryos of aegilops tauschii are selected as explants, materials are easy to obtain, the cost is low, and a large number of calluses can be produced by single seed; the method has the advantages of simple flow, strong repeatability and high differentiation rate, and lays a stable platform for genetic transformation and gene editing.

A tetraploid thinopyrum ponticum salt-tolerant 3el chromosome molecular marker and application thereof

The application discloses a tetraploid long-spike Aegilops tauschii salt-tolerant 3EL chromosome molecular marker and application thereof, and belongs to the technical field of crop genetic breeding. LTTE3EM-2 and LTTE3EZ-1 ) and a specific molecular marker of a long arm of a 6A chromosome of common wheat ( LTTAE6A-2 ) are combined into two groups of molecular markers ( LTTE3EM-2 and LTTAE6A-2, LTTE3EZ-1 and LTTAE6A-2 ), and a primer pair is designed based on the two groups of molecular markers, and it is found through verification that the designed primer pair can be used for identifying the genotype of a salt-tolerant wheat-tetraploid long-spike Aegilops tauschii 3EL translocation line, that is, whether the tetraploid long-spike Aegilops tauschii salt-tolerant 3EL chromosome is homozygous or not can be identified. The application provides an important way for wheat tribe system evolution, germplasm resource identification and molecular marker assisted selection breeding.
