Abstract

The invention discloses an in-vitro amplification method of northern pig-tailed macaque T cells, and belongs to the field of biotechnology. A commercialized non-human primate T cell activation / amplification kit is used for in vitro amplification of northern pig-tailed macaque T cells; the kit comprises magnetic beads and a biotinylated antibody, can simulate the effect of in-vivo antigen presenting cells, provides a first signal and a second signal for T cell activation, and effectively activates and purifies resting T cells in peripheral blood mononuclear cells. CD3 molecules are important markers on the surfaces of T cells and are combined with TCR through non-covalent bonds to form a TCR-CD3 complex, activation signals are transmitted into the cells, and activation and proliferation ofT cells are induced. CD28-mediated co-stimulatory signals can induce differentiation and proliferation of T cells and secretion of cytokines such as IL-2 and the like.

Description

technical field [0001] The invention relates to the field of biotechnology, in particular to an in vitro expansion method of T cells of the northern flat top monkey. Background technique [0002] Acquired immunodeficiency syndrome (AIDS) is an infectious disease caused by human immunodeficiency virus type I (HIV-1). HIV-1 virus mainly damages the body's immune system and seriously threatens human life and health. For a long time, in order to conquer HIV-1 virus infection, researchers have made many efforts. The non-human primates (NHP) model is an ideal animal model for studying the pathogenesis, vaccines and drugs of AIDS. In previous studies, primate surrogate models infected with simian immunodeficiency virus (SIV) or simian / human immunodeficiency virus (SHIV) were the most widely used HIV / AIDS animal models. However, the gene homology between SIV and SHIV virus and HIV-1 virus is quite different, which cannot truly reflect the clinical immune response of the body infe...
