[0020] 1. Isolation and cloning of cotton GhFEM1 genome DNA sequence and cDNA

[0021] Cotton fiber RNA was extracted 9 days after flowering by CTAB method (performed by Li XB, Cai L, Cheng NH, Liu JW, 2002.Molecular characterization of the cotton GhTUB1 gene that preferentially expressed in fiber.Plant Physiol.130:666-674), with SuperScript TM II RNase H-Reverse Transcriptase (Invitrogen Life Technologies) was reverse transcribed into cDNA; then, using cDNA as template, gene-specific primers (GhFEM1-ORF-L 5'-GGGGAATTCATGGATGTAACAAGCACACC-3' and GhFEM1-ORF-R 5' -CTTGGATCCTCACTCGATCTTACTCATCC-3') and high-fidelity DNA polymerase PFU, 94°C-5 min, 1 cycle; 94°C-1 min, 58°C-1 min, 72°C-1.5 min, 29 cycles; finally, 72 Under the PCR reaction conditions of elongation at ℃ for 10 minutes, a DNA fragment of about 1 kb (FEM1 gene) was obtained by amplification, as shown in SEQ ID No. 1. This fragment was inserted into the pBluescript SK-vector. Use the T3 and T7 promoters on the pla...

Abstract

The invention relates to a MYB gene for regulating the elongation and growth of cotton fibers and its application. The gene is derived from Gossypium hirsutum, and its full-length nucleotide sequence is 1149 bp, without introns, and its open reading frame is 933 bp, encoding a protein with 310 amino acids. This transcription factor has a typical MYB domain and a transcriptional repressor-binding domain (EAR-domain). The present invention verifies the function of the gene in regulating the elongation and development of cotton fibers through genetic transformation. Using RNAi technology to reduce the expression of the gene in cotton fibers can significantly reduce the length of cotton fibers, indicating that the gene is a positive regulator of cotton fiber elongation. The long-developed genes are of great value in the improvement of cotton fiber quality, and can be used for molecular breeding research on cotton fiber quality improvement through hybridization, transgenic and other methods.

Description

Technical field: [0001] The present invention relates to a MYB family transcription factor which is predominantly expressed in the rapid elongation period of cotton fiber and its encoding gene, named GhFEM1 ( F iber E longation-related M YB transcription factor 1), which can positively regulate the elongation and growth of upland cotton fibers. The FEM1 gene involved in the present invention is a gene with important value in cotton fiber quality improvement, and the gene can be used for molecular breeding research of cotton fiber quality improvement through technical methods such as hybridization and transgenic. Background technique: [0002] Cotton (Gossypium hirsutum) is an important raw material and strategic material reserve resource for the cotton textile industry. The cotton industry plays a pivotal role in the production of my country's national economy. At present, there is a shortage of domestic "high-grade cotton" and an excess of "low-grade cotton" in China, wh...
